摘要
以3个不同成熟度伽师瓜果实为试材,采用液相氧电极法、紫外分光光度计法分别测定总呼吸速率、5种途径呼吸速率及7种呼吸代谢关键酶的活性,通过分析不同成熟度伽师瓜果实采后各途径呼吸速率及其相关酶活性大小的关系,了解成熟度对伽师瓜采后呼吸的影响,确定适宜的采收成熟度。结果表明,成熟度2伽师瓜采后呼吸高峰出现比成熟度1和成熟度3伽师瓜晚20 d。成熟度2伽师瓜采后呼吸跃变过程以三羧酸循环途径的呼吸速率对总呼吸速率的贡献相对最高,三羧酸循环、细胞色素、交替途径的呼吸速率分别与琥珀酸脱氢酶、细胞色素氧化酶、交替氧化酶活性呈极显著正相关(P<0.01)。成熟度1伽师瓜采后的呼吸跃变过程以交替途径的呼吸速率对总呼吸速率的贡献相对最高,糖酵解和交替途径的呼吸速率分别与丙酮酸激酶、交替氧化酶活性呈极显著正相关(P<0.01)。成熟度3伽师瓜采后的呼吸跃变过程以细胞色素途径的呼吸速率对总呼吸速率的贡献相对最高。综合比较,选取成熟度2(10%<可溶性固形物含量≤12%)为伽师瓜果实适宜贮运的采收成熟度。
Through analyzing the relationship of postharvest respiration rate of Jiashi melon with different maturity and related enzyme activity, the effects of maturity on postharvest breathing of Jiashi melon, the proper maturity stage to harvest were explored and determined. The respiration rates of total respiration and 5 respiratory pathway of the Jiashi melons and the activities of 7 key enzymes involved in respiration were determined by the oxytherm system and ultraviolet spectrophotometer during storage.The results showed that,the postharvest respiration peak of Jiashi melon harvested in maturity stage 2 appeared 20 days later than maturity stage 1 and maturity stage 3,and its TCA pathway respiration rate had a higher contribution for total respiration in climacteric, while the respiration rates of TCA, CP, AP pathway had a significantly positive correlation (P〈0.01) with activities of SDH,COX and AOX respectively. AP respiration rate of melon in maturity stage 1 had a higher contribution for total respiration in climacteric, while the respiration rates of EMP and AP pathway had a significantly positive correlation(P〈0.01) with activities of PK and AOX respectively. CP respiration rate of melon in maturity stage 3 had a higher contribution for total respiration in climacteric. In conclusion, Jiashi melon harvested in maturity stage 2 with 10%-12% SSC was proper for storage and transportation.
