不同海拔猪血液抗氧化指标与耳组织相关基因表达的差异分析 被引量:1

摘要 为了研究不同海拔对猪血液中抗氧化指标和耳组织中相关基因表达是否存在差异,试验选择具有明显海拔高度差异的迪庆藏猪(3 500 m)和保山猪(2 600 m)作为研究对象,分别采集5月龄迪庆藏猪、保山猪的血液和耳组织,检测血清中一氧化氮(NO)、丙二醛(MDA)含量以及总超氧化物歧化酶(T-SOD)、过氧化物酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)的活力,并检测耳组织中SOD、CAT和GSH-Px基因表达的变化。结果表明:两个品种猪血清中NO、MDA含量和GSH-Px的活力没有显著性差异。迪庆藏猪血清中CAT的活力显著高于保山猪(P<0.05);T-SOD的活力极显著高于保山猪(P<0.01);此外,迪庆藏猪耳组织中CAT的基因表达显著高于保山猪(P<0.05);SOD的基因表达水平极显著高于保山猪(P<0.01);GSH-Px的基因表达水平有显著高于保山猪的趋势(P=0.071)。说明一定的海拔高度能够使猪产生氧化应激,高海拔的猪抗氧化应激能力更强。 Aim of the current study is to determine whether there is a difference in the antioxidant index of pig blood and related genes expression of ear tissues at different altitudes. The Diqing Tibetan pig (3 500 m) and the Baoshan pig (2 600 m) were chosen as experimental animals. The blood and ear tissues was collected from Diqing Tibetan pig and Baoshan pig at five - month, respectively ; samples of the collected blood were subjected to detection of nitric oxide( NO), malondialdehyde (MDA), the activity of total super- oxide dismutase (T -SOD), peroxidaze (CAT) , and glutathione peroxidase ('GSH -Px) ; samples of the ear tissues were subjected to detection of the gene expression of SOD, CAT and GSH - Px. The results indicated that there was no significant difference in the content of NO, MDA and the activity of GSH - Px in samples of sera from the two different altitudes. However, the activity of T - SOD ( P 〈0. 01 ) and CAT ( P 〈0. 05 ) in Diqing Tibetan pig serum were remarkable higher than that of Baoshan pig. Moreover, the gene expression of SOD ( P 〈 0.01 ) and CAT ( P 〈 0.05 ) in Diqing Tibetan pig were extremely higher than Baoshan pig; there was a tendency that GSH- Px gene expression (P = 0. 071 ) of Diqing Tibetan pig are higher than Baoshan pig. These data suggest that a certain altitude may cause oxidative stress in pigs, and the ability against oxidative stress of the pig is more better at high altitude.
