维氏气单胞菌3种免疫原对鲫鱼ACP、AKP、POD和SOD活力的影响 被引量:6

The effects of three types of Aeromonas veronii immunogen on activities of ACP,AKP,POD and SOD in blood of Carassius

摘要 为了研究维氏气单胞菌3种免疫原对鲫鱼酸性磷酸酶(ACP)、碱性磷酸酶(AKP)、超氧化物歧化酶(SOD)和过氧化物酶(POD)活力的影响,制备维氏气单胞菌强毒株TH0426的甲醛灭活全菌(FKC)、胞外产物(ECPs)以及灭活全菌与胞外产物(FKC+ECPs)的混合物三种免疫原,以其免疫鲫鱼,在免疫后的7,14,21,28,35,42天分别取样,测定血清中ACP、AKP、SOD和POD活力。结果表明:免疫后四种酶活力均发生了变化。各免疫组ACP活力随时间延长升高并维持在稳定状态,FKC+ECPs组在28天达到峰值,ECPs组和FKC组在35天达到峰值;各免疫组AKP活力逐渐下降并趋于稳定;各免疫组SOD活力随时间延长先升高再下降,均在35天达到峰值;各免疫组POD活力同样随时间延长先升高再下降,在28天达到峰值。说明三种免疫原均可诱导鱼体产生非特异性免疫,而且这四种酶活力可作为评价鱼体免疫效果的指标。 In order to determine the effects of three types of Aeromonas veronii immunogen on activities of acid phosphatase (ACP), alkaline phos-phatase (AKP), snperoxide dismutase (SOD) and peroxidase (POD) in the Blood of Carassius, three types of immunogen were prepared from Aeromonas veroni TH0426, a virulent c carp isolate, respectively. Healthy Carassius were vaccinated by the three types of the prepared immunogen, involving in formalin - killed cells ( FKC), extracellular proteins ( ECPs), and mixture of the former two components ( FKC + ECPs). Samples of blood was tested at 7, 14,21,28, 35, 42 d after post - vaccination. The activities of ACP, AKP, SOD and POD were detected in samples of the blood which were collected from the caudal veins of fish. The results indicated that the activities of four enzymes showed changes after vaccination, and the activity of ACP in each group increased with time and remained stable. In the FKC + ECPs group, activity of ACP reached the peak value at 28 d after post - vaccination. In the FKC group and ECPs group, activity of ACP reached the peak value at 35 d after post - vaccination. the AKP activity of each immunized groups came down and stabilized; the SOD activities decreased gradually after vaccination and there were slight difference among four groups, and reached the peak value at 35 d after post -vaccination; after challenging, the POD activities came down in all groups, and reached the peak value at 35 d after post-vaccination. The change of four enzymes activities indicated that the three immuno- gens can induce non -specific immunity in Carassius, and ACP,AKP, SOD and POD is an effective indicators of fish immune.
