染料木黄酮与大豆苷元对氨致星形胶质细胞水肿的抑制作用比较 被引量:1

摘要 目的对比研究染料木黄酮和大豆苷元对氨致星形胶质细胞水肿的抑制作用。方法以MTT法检测细胞活力,以3-O-甲基-[~3H]-葡萄糖检测细胞水肿。以蛋白质免疫共沉淀法检测ERK/EGFR相互作用。结果 50μmol/L染料木黄酮和大豆苷元均不影响细胞活力。染料木黄酮能显著抑制氨引起的细胞水肿和ERK/EGFR相互作用,而大豆苷元则无此生物学活性。结论染料木黄酮有显著的抗星形胶质细胞水肿活性,这可能与其特有的EGFR抑制活性有关。 Objective To compare the inhibition of genistein and daidzein on ammonia-induced astrocyte swelling.Methods Cell viability was examined with MTT assay,cell swelling with3-O-methyl-[3H]-glucose.Co-immunoprecipitation was used to determine ERK/EGFR interaction.Results Genistein and daidzein at 50 μmol/L had no effect on astrocyte viability.Genistein,but not daidzein,produced significant inhibitory effect on ammonia-induced astrocyte swelling and ERK/EGFR interaction.Conclusion Genistein exhibits a significant inhibitory effect on ammonia-induced astrocyte swelling,which may be related to its specific inhibition on EGFR.
