摘要
目的通过检测系统性红斑狼疮(SLE)人群和正常对照人群SOX5基因表达的差异,探讨其在SLE发病中的意义。方法收集20例系统性红斑狼疮患者和15例健康体检者(对照组)的外周血和临床资料。采用实时荧光定量聚合酶链式反应(Quantitative Real-time PCR)检测各个样本外周血单个核细胞中SOX5mRNA和IL-17mRNA的表达水平,计算SLE组和正常对照组的SOX5mRNA表达水平的差异(以-△△CT值表示)。结果 SOX5mRNA在SLE组的表达较健康对照组的表达显著增高(P<0.05),且SOX5mRNA和IL-17mRNA有一定的相关性(Spearman r=0.45,P<0.05)。结论 SOX5可能作为炎性反应的正向调控基因参与了系统性红斑狼疮的免疫反应过程。
Objective To compare the expression of SOX5 in systemic lupus erythematosus (SLE) patients and normal individuals, and to investigate its role in the development of SLE. Methods The peripheral blood and clinical data of 20 cases of SLE and 15 cases of healthy cases were collected. Quantitative real-time PCR was used to detect the expressions of SOX5mRNA and IL-17mRNA in mononuclear cells in the sample pcripheral blood and the expression levels(indicated as-△△CT value)of SOX5 in the SLE patients and the controls were calcfflated. Results The expression level of SOX5mRNA in the SLE group was significantly higher than that in the healthy group ( P 〈0.05), and there was a correlation between the expression levels of SOX5 mRNA and IL-17 mRNA ( Spearman r =0.45, P 〈0.05). Conclusion SOX5 may act as the positive regfflatory genes of inflammatory reaction and may be involved in the immune response in SLE.
