摘要
目的通过检测维甲酸相关孤核受体(RORC2)在系统性红斑狼疮(SLE)患者外周静脉血单个核细胞(PBMCs)中的表达,初步探讨RORC2在SLE免疫炎症反应中发挥作用的机理。方法选取SLE患者21例及15名健康体检者为对照组,分离外周静脉血单个核细胞(PBMCs),提取总RNA作模板,采用逆转录-聚合酶链反应(PT-PCR)检测SLE患者及健康对照组Th17细胞转录因子RORC2和IL-17的mRNA的表达水平并进行组间比较。结果与正常人对照组相比,SLE组的RORC2mRNA表达显著提高,两组差别有统计学意义(P<0.05);SLE组的RORC2和IL-17mRNA表达水平也存在着相关性。结论 RORC2通过调控Th17细胞相关效应因子IL-17的分泌,促进T细胞免疫炎症反应。进一步提示,通过阻断Th17细胞分化的上游活动来减轻SLE的免疫炎症反应可以达到控制SLE的目的。
Objective To explore the mechanism of RORC2 in the response of systemic lupus erythematosus(SLE)by detecting the expressions of RORC2 in the PBMCs of SLE patients.Methods 21 cases of SLE and 15 healthy cases were chosen to separate PBMCs and to extract the total RNA as the templates.The mRNA expression levels of RORC2 and that of IL-17 were detected by reverse transcription-polymerase chain reaction(RT-PCR)and analyzed.Results The RORC2 mRNA expression level of SLE patients was significant higher than that of the normal control group(P 〈0.05).There existed correlation between the expression level of RORC2 mRNA and that of IL-17 mRNA in SLE patients.ConclusionBy regulating the IL-17 secretion of Th17 cells,RORC2can promote T lymphocyte cell immune reaction,which further proves that cutting the upstream activities of Th17 cell differentiation can control the development of SLE diseases.
