下调Nek2表达对肾癌细胞侵袭、迁移的影响 被引量:3

摘要 目的观察下调中心体相关激酶2(Nek2)对肾癌细胞侵袭及迁移的影响,探讨Nek2在肾癌细胞侵袭与迁移中的作用。方法培养人肾癌细胞系A498细胞,分为观察组、对照组,分别给予100 mmol/L的Nek2抑制剂INH6及等体积DMSO,均培养14 h。采用q-RT-PCR法检测Nek2 mRNA表达,采用Western blot法检测Nek2蛋白表达。采用Transwell细胞侵袭实验测定细胞侵袭能力,采用细胞划痕实验测定细胞迁移能力。结果对照组、观察组Nek2 mRNA表达量分别为1.00±0.01、0.22±0.01,观察组Nek2 mRNA表达量低于对照组(P<0.01)。对照组、观察组Nek2蛋白表达量分别为1.56±0.12、0.56±0.10,观察组Nek2蛋白表达量低于对照组(P<0.01)。对照组、观察组侵袭细胞数分别为(428.8±18.1)、(128.2±46.9)个,观察组侵袭细胞数低于对照组(P<0.01)。对照组、观察组划痕愈合率分别为86.3%±5.7%、53.2%±3.4%,观察组划痕愈合率低于对照组(P<0.01)。结论下调Nek2的表达可抑制肾癌细胞的迁移和侵袭,Nek2可能参与了肾癌迁移与侵袭的过程。
