摘要
为研究浓缩Vero细胞狂犬病疫苗蛋白浓度对纯化效果的影响 ,用超滤浓缩法对狂犬病疫苗进行不同倍数的超滤浓缩 ,将浓缩后不同倍数的样品分别用凝胶柱层析法进行纯化。结果样品浓度在 4 0mg/ml以下时 ,狂犬病毒收集液中杂蛋白去除率可达 99%以上 ,小牛血清含量在 2 5~ 37.5ng/ml之间 ;浓度超过 4 0mg/ml时 ,并随着样品浓度的增加 ,狂犬病毒收集液中杂蛋白去除率逐渐下降 ,且有病毒丢失。因此采用凝胶柱层析法纯化狂犬病疫苗 ,蛋白浓度应低于 4 0mg/ml。
For investigating the influence of sample protein concentration upon the purification effect of rabies vaccine (vero cell culure) during process of gel column chromatography, the starting material of rabies vaccine was ultrafiltrated to produce preparations of different concetrated folds, which are then purified respectively with gel column chromatography. The resellts show the removing rate of impure protein in harvest of rabies virus reached more than 99% and the residual protein content of calf serum became between 25~37.5 hg/ml when the protein concentration of applied sample is lower than 40mg/ml; but the removing rate of impure protein in harvest of rabies virus decreased gradually and a certain amount of virus lost when applied sample concentration is higher than 40mg/ml.Therefore, the protein concentration of applied sample should be less than 40 mg/ml when the rabies vaccine is purified with gel column chromatography.
