结扎腰动脉建立大鼠脊髓缺血模型有效性的评估

摘要 目的结扎腰动脉建立大鼠脊髓缺血模型,为研究缺血性脊髓病、继发性脊髓损伤的发生机制提供良好的载体。方法将大鼠随机分为假手术组(仅分离出腰动脉)和脊髓缺血组(结扎所有腰动脉),于术后进行大鼠后肢运动功能BBB评分、脊髓苏木精-伊红(H-E)染色镜下观察及Caspase-3免疫组织化学检测。结果术后48h对缺血组大鼠后肢运动功能BBB评分为(9.22±0.97),与对照组比较,差别有统计学意义(P<0.01);H-E染色镜下可见脊髓缺血组大鼠正常神经元数目明显减少;免疫组织化学显示Caspase-3阳性细胞呈棕黄色,脊髓缺血组阳性率远大于假手术组,差别有统计学意义(P<0.01)。结论通过结扎大鼠腰动脉可以建立稳定的脊髓缺血模型,为脊髓缺血后损伤机制和预防措施的研究提供良好的载体。 Objective To evaluate a mouse model produced by ligation of the lumbar arteries for study of spinal cord diseases and spinal cord injury.Methods SD rats were divided into the sham operation group and spinal cord ischemia group.In the spinal cord ischemia group the lumbar arteries were ligated while in the sham operation group we only separated out all lumbar arteries.BBB scoring of motor function of hindlimb of rats,the hematoxylin-eosin staining(H-E)and immunohistochemistry test were performed after the vascular ligation or the separation of the arteries.Results BBB score of motor function of hind limb of rats 48 hours after vascular ligation was(9.22±0.97),which was statistically significantly different than the BBB score in the sham operation(P〈0.01).The result of spinal cord ischemia HE was that the number of normal neuron decreased.Immunohistochemistry showed that the sham operation group had only a few scattered yellow-brown positive cells of Caspase-3,while the spinal cord ischemia group showed numerous and densely distributed positive cells,and the difference was statistically significant(P〈0.01).Conclusion By ligating the lumbar arteries in the rats,we can make stable spinal cord ischemia models therefore are able to supply an ideal model for spinal cord diseases and spinal cord injury research.
