摘要
目的:探讨实验室合成的2-甲氧基雌二醇(2-ME)及其合成中间体的大鼠肺细胞膜色谱行为。方法:经筛选,大鼠细胞膜色谱条件为自制大鼠肺细胞膜色谱柱(10 mm×2 mm),流动相为10 mmol/L磷酸盐缓冲液(p H7.4),柱温为37℃,流速为0.2 m L/min。以沙丁胺醇为阳性对照,以容量因子k'作为指标,考察2-ME及其合成中间体在大鼠肺细胞膜色谱柱上的保留特性。结果:2-ME及合成中间体1、2号化合物在大鼠肺细胞膜色谱柱上均有吸附,且k'依次为2号化合物>1号化合物>2-ME;合成中间体3及4号化合物未表现出吸附特性。2-ME与大鼠肺细胞膜色谱柱的亲和力大于纯硅胶色谱柱、大鼠心肌细胞膜色谱柱和肝细胞膜色谱柱。结论:2-ME在大鼠肺细胞膜上具有特异性亲和。
Aim: To screen the activity of 2-methoxyestradiol( 2-ME) and synthetic derivatives by rat lung cell membrane chromatography. Methods: The lung cell membrane stationary phase( CMSP) was packed into a stainless steel column( 10 mm × 2 mm),and the HPLC conditions were as follows: a mobile phase consisted of 10 mmol / L phosphate buffer( p H 7. 4) at a flow rate of 0. 2 m L/min,and the column temperature was 37 ℃. Salbutamol was used as positive control in the screening. The capacity factor k' as chromatographic parameters was used to describe the affinity of 2-ME and its synthetic derivatives. Results: 2-ME,NO. 1 synthetic derivative and NO. 2 synthetic derivative had chromatographic retention on lung CMSP column and k' of NO. 2 was greater than NO. 1 and 2-ME. The k' of 2-ME with rat lung CMSP was higher than those with pure silica gel column,rat cardiac muscle CMSP and liver CMSP. Conclusion: 2-ME has the property of specific affinity on the membrane of the rat lung cell membrane.
