摘要
通过研究贵州三穗鸭损伤表皮调节因子(Bmal1)基因的多态性,寻找其SNP多态位点及分析其在组织中的表达差异。利用PCR-SSCP技术与直接测序技术相结合和荧光定量表达的方法对Bmal1基因进行SNPs位点筛选和遗传特性分析,根据测序结果拼接获得的序列对该基因mRNA相关生物信息学进行分析。贵州三穗鸭Bmal1基因第一外显子存在两处SNP位点,且在脑组织中的表达量最高,2^(-△△CT)值为1.4431。Bmal1基因突变前后的等位基因频率估算有差异,但其mRNA二级结构预测,蛋白质二、三级结构预测分析均无差异。在心、肝、肾、胸肌、腿肌、脑、眼七个组织中表达存在差异,且各组间差异显著。
In order to study the polymorphisms of Bmall gene, find its SNP polymorphic sites and analyze its expression in different tissues of Guizhou Sansui duck, PCR-SSCP technique,direct sequencing technology, qRT-PCR technology and SPSS 19.0 software were combined to select the SNP loci of Bmall gene in Sansui duck,analyze its genetic effects,predict the secondary structure of mRNA and secondary or tertiary structure of protein. In this experiment, two SNP loci of the exon 1 in Guizhou Sansui duck Bmall gene were found. The expression level of Bmall gene in the brain was the highest. Additionally,Allele frequencies of gene were differences before and after mutation. No differences were found when predicting its mRNA secondary structure,protein secondary and tertiary structure. There were significant differences among the seven tissues of heart,liver,kidney,breast muscle,leg muscle,brain and eye.
