mTOR在人脂肪源间充质干细胞成脂中的作用

摘要 通过探讨mTOR对hADSCs成脂的影响,为脂肪分化异常所致疾病的预防和治疗提供参考.针对通过吸脂术所抽取的人腹部脂肪组织,利用Ⅰ型胶原酶消化分离培养hADSCs,三联诱导法使其向脂肪细胞分化,油红O染色鉴定.通过免疫细胞化学染色法检测mTOR在细胞内的分布变化;观察雷帕霉素抑制mTOR后hADSCs成脂能力的变化,即油红O染色及PPARγ的表达.结果表明,所分离细胞为平行排列、生长形态相对均一的梭形细胞.三联诱导法诱导7d后可见脂滴的出现,油红O染色着红色.mTOR的分布在分化过程中由核内转向胞浆.雷帕霉素处理细胞使细胞内脂滴的蓄积显著减少、PPARγ表达降低.雷帕霉素抑制mTOR表达可降低hADSCs的成脂能力. By studying the influence of mTOR on hADSCs adipogenesis,the article provides reference for prevention and treatment of adipocyte prosoplasia related diseases.Specific to the human abdominal adipose tissues extracted by liposuction,Ⅰ collagenase was used to digest and seperate hADSCs,triple inducement method made the differentiation of adipose adipocyte,and oil red O staining identify.The result shows that,the isolated cells are fusiform cell with parallel arrangement and comparatively even in growth form.7days after inducing via triple induction method shows visible lipid drops,and oil red O redcolored.The distribution of mTOR turns from intranuclear to cytoplasm during the process of atomization.The processing of cells by sirolimus made the accumulation of lipid droplet in cells reduce significantly and the express of PP PPARγdecrease.The restraining of sirolimus on mTOR express can decrease the adipogenesis ability of hADSCs.
