克拉霉素对COPD大鼠的抗炎作用

摘要 目的探讨克拉霉素对COPD大鼠的抗炎效果及对肺功能的改善情况。方法建立COPD大鼠模型,实验分为健康对照组、COPD组与克拉霉素组,每组10只。实验结束后检测大鼠肺功能,留取血液及肺泡灌洗液,使用ELISA法检测血清及肺泡灌洗液中INF-γ、TNF-α、NF-κB的水平。结果与对照组相比,COPD组大鼠,0.3 s最大呼气量/用力肺活量(forced expiratory volume 0.3/forced vital capacity,FEV0.3/FVC)明显低于对照组,差异具有统计学意义(P<0.05);吸气阻力(inspiratory resistance,Ri)、呼气阻力(expiratory resistance,Re)明显高于对照组,差异具有统计学意义(P<0.05)。克拉霉素组FEV0.3/FVC高于COPD组,Ri及Re明显低于COPD组,差异具有统计学意义(P<0.05)。COPD组血清及肺泡灌洗液中INF-γ、TNF-α、NF-κB的水平明显高于对照组,差异均具有统计学意义(P<0.05);克拉霉素组血清及肺泡灌洗液INF-γ、TNF-α、NF-κB的水平明显低于COPD组,差异均具有统计学意义(P<0.05)。结论克拉霉素能在一定程度上减轻COPD大鼠肺部及全身的炎症反应,改善COPD大鼠肺功能。 Objective To investigate the anti - inflammatory effect of clarithromycin on chronic obstructive pulmonary disea- ses (COPD) rats and the improvement of pulmonary function. Methods COPD rat model was established. Rats were randomly divided into 3 groups: control group, COPD group and elarithromycin group, 10 rats in each group. After the experiment, the puhnonary function of rats was determinated, and the serum and alveolar lavage fluid was collected for detecting the levels of INF-γ、TNF-α、NF-κB. Results Compared with control group, the expiratory forced capacity volume 0.3/forced vital ca- pacity (FEV0.3/FVC) , resistance inspiratory (Ri) and resistance expiratory (Re) in the COPD group were significantly lower than blank group, and the differences were statistically significant (P 〈 0. 05 ). Compared with COPD group, FEV0.3/FVC, Ri and Re in the clarithromyein group were higher than group COPD, and the differences were statistically significant ( P 〈0. 05 ). In COPD group, INF-γ、TNF-α、NF-κB level in serum and alveolar lavage fluid were higher than those in the blank group, and the differences were statistically significant ( P 〈 0. 05 ). In clarithromycin group, the levels ofINF-γ、TNF-α、NF-κB in serum and alveolar lavage fluid were lower than group COPD, and the differences were statistically significant ( P 〈0. 05 ). Conclusion Clarithromycin can reduce the pulmonary and systemic inflammation in COPD rat to a certain extent, and improve the pulmonary function.
