加味茵陈四逆汤对肝纤维化小鼠TGF-β_1/Smads通路的影响 被引量:15

Effect of Jiawei Yinchen Sini Decoction on the TGF-β_1/Smads Signal Pathway in Mice with Hepatic Fibrosis

摘要 目的:探讨加味茵陈四逆汤预防肝纤维化过程中对TGF-β_1/Smads通路的影响。方法:将48只雄性ICR小鼠随机分为正常对照组、模型组、秋水仙碱(0.1 g/kg)阳性对照组及加味茵陈四逆汤高(41.6 g/kg)、中(20.8 g/kg)、低(10.4 g/kg)剂量组,每组8只。除正常对照组外,其余各组采用腹腔注射30%CCl4复制肝纤维化模型,造模同时给予相应药物,正常对照组与模型组给予等体积蒸馏水。给药14 d后,比较小鼠体质量和肝脏组织病理学改变;试剂盒检测血清ALT、AST水平;采用RT-q PCR检测肝组织TGF-β_1、TβRⅠ、Smad3 mRNA表达。结果:与正常对照组比较,模型组大鼠体质量显著降低,血清ALT、AST水平显著升高,肝组织呈明显的纤维化改变,肝组织TGF-β_1、TβRⅠ、Smad3 mRNA表达显著升高。加味茵陈四逆汤能不同程度提高模型小鼠体质量,降低血清ALT、AST水平,改善肝组织病理学改变,并降低肝组织TβRⅠ、Smad3 mRNA表达。结论:加味茵陈四逆汤能预防CCl4诱导的肝纤维化,其作用机制可能与下调肝脏组织TβRⅠ、Smad3 mRNA的表达,从而阻滞TGF-β_1/Smads通路转导有关。 Objective: To explore the mechanism of Jiawei Yinchen Sini decoction in preventing hepatic fibrotic mice via TGF-β1/Smads signal pathway. Methods: 48 male ICR mice were randomly divided into normal control group,model group,colchicine positive control group,Jiawei Yinchen Sini decoction high dose group( 41. 6 g / kg),medium dose group( 20. 8 g / kg) and low dose group( 10. 4g / kg),with eight mice in each group. Except for the normal control group,the mice were treated with intraperitoneal injection of 30%CCl4to estabish hepatic fibrosis model,and the mice were treated with intragastric adminstration of corresponding drugs,but in the normal control group and model group were treated with equal volume distilled water. After 14 days,the body weight was calculated and the pathological hepatic fibrosis changes in liver of mice were observed. Levels of serum alanine transarninase( ALT),aspartate transarninase( AST) were detected by assay kits. The expression of TGF-β1,TβRⅠ and Smad3 mRNA in liver tissues were detected by RT-q PCR analysis. Results: Compared with normal control group,the body weight in model group was significantly reduced,the levels of serum ALT and AST were significantly higher,pathological hepatic fibrosis were changed remarkable. The expression levels of TGF-β1,TβRⅠand Smad3 mRNA were higher. Jiawei Yinchen Sini decoction could increase different levels of body weight in model mice,and decreased the levels of ALT and AST in serum,improved the pathological hepatic changes and decreased the expression levels of TβRⅠand Smad3 mRNA. Conclusion: Jiawei Yinchen Sini decoction can prevent hepatic fibrotic mice,and the mechanism of Jiawei Yinchen Sini decoction in preventing hepatic fibrosis induced by CCl4 could be related to the inhibition of TβRⅠ mRNA expression and Smad3 mRNA expression and blocking of TGF-β1/ Smads signal pathway.
