摘要
目的:研究湖南产乌药化学成分。方法:分别采用气相色谱-质谱(GC-MS)联用法和液相色谱-质谱(UPLC-ESI-MS/MS)对乌药中挥发油和甲醇提取物的化学成分进行分析。在GC-MS中,以HP-5MS毛细管柱(30 m×0.25 mm,0.25μm)为色谱柱,采用程序升温法,EI离子源,扫描质量范围为m/z 35~400 amu;在UPLC-ESI-MS/MS中,以Acquity UPLC BH C18(50 mm×2.1 mm,1.7μm)为色谱柱,采用0.1%甲酸水溶液-乙腈为流动相,梯度洗脱,ESI源,扫描质量范围为m/z 100~1 200。结果:利用GC-MS法共鉴定了乌药中68个挥发性成分,主要为单萜类物质。利用UPLC-ESI-MS/MS法在甲醇提取物中共鉴定出42个化合物,其中24个呋喃倍半萜及其内酯、10个生物碱、6个黄烷醇多酚及其缩合物、1个有机酸和1个甾醇类化合物。结论:系统研究了湖南产乌药中的化学成分,为湖南产乌药的药效物质基础及质量控制研究提供一定的理论依据。
Objective: To study the chemical composition of Lindera aggregate produced in Hunan. Methods: Volatile oil of Lindera aggregata and chemical composition of methanol extract were analyzed by GC-MS and UPLC-ESI-MS / MS. The GC-MS condition was as follows,HP-5MS capillary column( 30 m × 0. 25 mm,0. 25 μm) for column chromatography,temperature-programmed and EI were used,the scanning range of quality was m / z 35 ~ 400 amu; the UPLC-ESI-MS / MS conditions were as follows,Acquity UPLC BHC18( 50 mm ×2. 1 mm,1. 7 μm) with mobile phase of 0. 1% formic acid aqueous solution and acetonitrile under gradient elution,with the EI source. The scanning range was m / z 100 ~ 1 200. Results: 68 volatile compounds were identified in Lindera aggregata,mainly monoterpene substance,42 compounds were identified,24 furan sesquiterpene and lactone,ten alkaloids,six flavonol polyphenol and condensates,an organic acid and a sterol classes of compounds in the methanol extract of Lindera aggregata. Conclusion: This study make a comprehensive understanding of compounds of Lindera aggregata which produced in Hunan,and provides a theoretical basis for pharmacodynamic substance foundation and the quality control of Lindera aggregata.
