AuNCs@SiO_2金纳米簇复合粒子制备及其对体外口腔癌细胞毒性的研究 被引量:2

摘要 目的制备一种具有荧光性的二氧化硅包覆金纳米簇复合粒子,并研究其体外细胞毒性。方法以牛血清白蛋白(BSA)介导合成金纳米簇(BSA-AuNCs),改良St9ber法制备二氧化硅包覆的金纳米簇复合粒子AuNCs@SiO_2并表征,通过CCK-8细胞相容性实验测试复合纳米粒子对人舌鳞癌细胞CAL27,人口腔表皮样癌细胞KB,小鼠胚胎成纤维细胞3T3的细胞毒作用。结果 AuNCs@SiO_2复合金纳米簇粒子在365nm紫外灯下具有荧光性,透射电镜显示纳米复合粒子为球形、粒度均匀的壳核型粒子,平均粒径52.30nm;紫外可见光吸收曲线显示,与金纳米簇相比较,氧化硅包覆的金纳米簇复合粒子特征性吸收峰消失,荧光光谱显示最大发射峰轻微蓝移至655nm处;CCK-8法结果显示,不同浓度AuNCs@SiO_2分别作用于细胞24h,各剂量组与正常对照组相比较,细胞存活率差异不具有统计学意义(P>0.05)。结论合成的金纳米簇壳核复合粒子不仅保留了金纳米簇的荧光特性,表层的硅壳还能够提高金纳米簇的荧光稳定性,并且纳米复合粒子对体外口腔癌细胞和小鼠3T3细胞无显著细胞毒性。此种金纳米簇复合粒子有望应用于口腔癌细胞的成像和治疗领域。 Objective To prepare fluorescent silica-coated gold nanoclusters nanocomposite particles and evaluate the cytotoxicity in vitro.Methods Silica coated gold nanoclusters(AuNCs@SiO2)was prepared by modified Stober method and characterized by the transmission electron microscope,UV-VIS spectrophotometer,fluorescence spectrophotometer etc.The cytotoxicity on CAL27,KB and 3T3 cells in vitro was assessed by CCK-8 method.Results The AuNCs@SiO2nanocomposite particles emit red fluorescence under 365 nm UV light;TEM showed the mean effective diameter of the core-shell nanoparticles was approximately 52.30 nm with uniform,spherical shape.The absorption peak of the AuNCs@SiO2disappeared in UV-vis spectra;The fluorescence spectrum showed that the emission peak exhibited a slight blue shift to 655nm;CCK-8assay showed that after cultured with different concentration of AuNCs@SiO2for 24 hours,the survival rates of three kind of cells had no statistical difference compared with the control group.Conclusion The silica layer played protective role to the photostability of the fluorescent AuNCs,and the nanocomposite particles had no obvious cytotoxicity on oral cancer cells CAL27,KB and mouse embryo fiberblasts3T3.AuNCs@SiO2nanomaterial is promising in malignant neoplasm cell image including oral cancer.
