混合纤维素微孔滤膜用于液体藻类的冷冻扫描电镜样品制备 被引量:8

摘要 为了优化针对液体培养藻类细胞的冷冻扫描电镜制样条件,以铜绿微囊藻和蛋白核小球藻为材料,比较了混合纤维素微孔滤膜(混合膜)和滤纸对藻细胞的富集作用以及冷冻扫描电镜观察效果。结果表明:混合膜的孔隙较小且孔径分布均匀,正反两面均能有效吸附藻细胞。与滤纸相比,混合膜上吸附的藻细胞数量较多,细胞形态饱满,无皱缩变形,成像效果好。混合膜比滤纸更适合于微小藻类液体样品的冷冻扫描电镜样品制备。 To optimize the sample preparation conditions of Cryo-scanning electron microscopy( Cryo-SEM),we compared the enrichment and Cryo-SEM observation effects of mixed cellulose ester membrane( MCEM) and filter paper for liquid algae cells.Microcystis aeruginosa and Chlorella( diameters about 2 to 3 μm) were chosen as materials. The pores of MCEM were small and uniform in distribution. Both front and back surfaces of MCEM could effectively adsorb algae cells. MCEM could adsorb more cells and get better imaging effects than the filter paper. Cryo-SEM images showed that cells on MCEM were plump in morphology,and with no distortion or shrinkage. MCEM was more suitable for micro liquid algae samples preparation for Cryo-SEM.
