摘要
目的:研究蒙古扁桃石油醚提取物对四氯化碳(CCl4)致大鼠肝纤维化的保护作用。方法:SD大鼠按体重随机分为对照组、模型组、阳性药水飞蓟素组(0.05 g·kg-1)和蒙古扁桃石油醚提取物高剂量组(1.75g·kg-1)、中剂量组(1.25 g·kg-1)、低剂量组(0.75 g·kg-1)。除对照组外,其余各组大鼠均皮下注射40%CCl4橄榄油溶液3 m L·kg-1(首次为5 m L·kg-1)每周2次,连续8周,造模同时各给药组灌胃给药。8周后测定丙氨酸氨基转移酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)、丙二醛(MDA)、超氧化物歧化酶(SOD)、肝纤维化指标透明质酸(HA)、层粘连蛋白(LN)、Ⅲ型前胶原(PCⅢ)、Ⅳ型胶原(CⅣ);HE、Masson染色观察病理学变化。结果:大鼠肝纤维化损伤时,模型组ALT、AST、ALP活性,血清HA、PCⅢ、CⅣ及MDA明显升高,而SOD明显降低,与对照组比较差异有显著性(P<0.05);与模型组比较,蒙古扁桃石油醚提取物可降低ALT、AST、ALP活性及血清HA、PCⅢ、MDA含量,而提高血清SOD含量,差异均有显著性(P<0.05)。病理学检查结果显示,蒙古扁桃石油醚提取物治疗组肝纤维化有显著改善。结论:蒙古扁桃石油醚提取物对四氯化碳所致的大鼠肝纤维化具有良好的保护作用。
Objective : To investigate the protective effect of Petroleum ether extract of Amygdalus mongolica on CC14 - induced hepatic fibrosis in rats. Method: Sixty Sprague- Dauley (SD) rats were randomly divided in groups ( one control group, one model group, one postitive silymarin controlled group, and three different doses (1.75 g · kg^-1 ,1.25 g · kg^-1,0.75 g· kg^-1) of petroleum ether extract of Amygdalus mongolica treatment groups. Liver fibrosis rat model was induced by carbon tetrachloride ( CCl, ). Except for control group, other groups were given drugs when being given CC14. At the End of the study (the eighth week), liver function (alanine amino transferase(ALT) ,aspartate amino transferase ( AST), alkaline phosphatase ( ALP ) ), malon dialdehyde ( MDA ), superoxide dismutase ( SOD ), hyaluronic acid ( HA ), laminin ( LN ) , procollagen - Ⅲ ( PC Ⅲ ) and collagen - Ⅳ ( C Ⅳ ) were measured. Pathological examination on liver tissue was also made. Result : Serum concentrations of ALT, AST, ALP, HA, PC Ⅲ, C Ⅳ, MDAwas significantly higher in the liver fibrosis model group, however, the concentrations of SOD were significantly lower in the model group. Compared with the model group, the serum concentrations of ALT, AST, ALP, HA, PC Ⅲ, MDA were significantly decreased in petroleum ether extract of Amygdalus mongolica treatment groups and the serum concentrations of SOD were significantly in- creased in petroleum ether extract of Amygdalus mongolica treatment groups. Pathological results showed that the liver fi- brosis in Petroleum ether extract of Amygdalus mongolica groups was significantly improved. Conclusion: Petroleum ether extract of Amygdalus mongolica has significantly protective effect on liver fibrosis induced by carbon tetrachloride in rats.
