摘要
目的研究山楂叶总黄酮(hawthorn leaves flavonoids,HLF)对高脂高盐所致高血压大鼠降压及心脏的保护作用,并探讨其作用机制。方法采用高脂高盐饲料加5%盐水连续喂养8周的方法建立高血压模型大鼠,给予不同剂量HLF[50、100和200 mg/(kg·d)]和卡托普利[25 mg/(kg·d)]进行干预治疗,疗程4周,并设模型组和正常对照组。实验前、造模后(给药前)及治疗完成后测量收缩压(systolic blood pressure,SBP)和舒张压(diastolic blood pressure,DBP);治疗完成后,测定血清中血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)水平、心肌增值系数(myocardial proliferation factor,MPI)及心脏做功系数(cardiac work index,CWI),测定血清中血脂监测指标[总胆固醇(total cholesterol,TC)、甘油三脂(triglycerides,TG)、低密度脂蛋白(low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白(high-density lipoprotein cholesterol,HDL-C)]水平和心肌酶[谷草转氨酶(aspartate transaminase,AST)、磷酸肌酸激酶(creatine phospho kinase,CPK)、乳酸脱氢酶(lactate dehydrogenase,LDH)]活性;测定心肌组织中抗氧化酶[超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、过氧化氢酶(catalase,CAT)]活性和丙二醛(malondialdehyde,MDA)含量。结果经HLF治疗4周能够有效降低高脂高盐所致高血压大鼠SBP和DBP,降低血清AngⅡ水平,降低MPI和CWI,降低血清中TC、TG、LDL-C含量并提高HDL-C含量,降低血清中AST、CPK、LDH活性,提高心肌组织中SOD、GSH-Px、CAT活性并降低MDA含量,且HLF上述作用均具有一定剂量依赖性。结论 HLF能够降低高脂高盐所致高血压大鼠血压并具有保护心脏的作用,其机制可能与HLF能够有效降低高脂高盐所致高血压大鼠AngⅡ水平、调节血脂、改善抗氧化酶活性、抑制氧化应激损伤有关。
Objective To investigate the effects of Hawthorn Leaves Favonoids( HLF) on blood pressure and myocardial tissue of hypertension rat induced by high fat and high salt. Methods 60 hypertension rats were fedded with high fat and high salt for eight weeks to induce high blood pressure model.The rats were treated with HLF[50,100,200 mg /( kg·d) ]and captopri [25 mg /( kg·d) ]for 4 weeks,and setting model group and normal control group. The SBP and DBP were determined before treatment,after modeling and after treatment. After 4 weeks,the level of Ang Ⅱ in serum and the MPI,CWI were determined; the content of TC,TG,LDL-C,HDL-C and the activity of AST,CPK,LDH in serum were determined; the activity of SOD,GSH-Px,CAT and the content of MDA in myocardial tissue were determined.Results The level of SBP and DBP in HLF treated groups were significantly decreased,the level of AngⅡ,MPI,CWI were significantly decreased; the content of TC,TG,LDL-C,AST,CPK,LDH in serum were significantly decreased while the HDL-C was significantly increased; the activity of SOD,GSH-Px,CAT in myocardial tissue were significantly increased and the content of MDA were significantly decreased;all of the effects has dose-dependent. Conclusions HLF can effectively decrease the blood pressure and protect the myocardial tissue,which is perhaps related with its pharmacological effects of cutting down the level of Ang Ⅱ of hypertension rat induced by high fat and high salt,reduce blood lipids,improve antioxidant enzyme activity and depress the oxidative stress.
