摘要
目的研究成都地区献血者人群HCV感染的基因型分布。方法从血液筛查出的抗-HCV反应性血清标本中提取HCV RNA,RT-PCR获得HCV核酸C区及NS5B区序列片段,用MEGA软件构建序列进化树,分析HCV的亚型分布特征及可能的原因。结果对313例HCV RNA阳性标本分型结果如下,1b亚型283例,2a亚型14例,3b亚型7例,3a亚型3例,6a亚型5例,6u亚型1例;4和5基因型没有发现。结论在成都地区献血者人群感染的HCV中共有6个亚型分布。其中1b是绝对优势亚型,占90.41%(283/313);6u亚型在成都地区首次被检测到。
Objective To study the HCV distribution characterization of genotypes from volunteer blood donors in Chengdu. Methods HCV RNA was extracted from samples with positive anti-HCV and HCV fragments and was amplified u- sing RT-PCR. Purified productions of PCR were sequenced and phylogenetic trees were constructed based on the HCV NS5B regions from 313 sequences. Results HCV genotypes were determined with the following results: lb in 283, 2a in 14, 3b in 7, 3a in 3, 6a in 5 and 6u in l. Genotype 4 and 5 were undetected. Conclusion Six HCV subtypes were found in blood do- nors from Chengdu area. Subtype lb was the most common and accounted for approximately 90. 41% (283/313), and one subtvoe 6u was firstly isolated from Chengdu area.
