医用臭氧对兔膝骨关节炎炎性因子水平的影响 被引量:8

Effect of medical ozone on the levels of inflammatory factors of knee osteoarthritis in rabbits

摘要 目的观察医用臭氧在兔膝骨关节炎中的应用及对关节滑液中肿瘤坏死因子(TNF-α)、白细胞介素(IL-1β)、基质金属蛋白酶-1(MMP-1)水平的影响。方法取新西兰大白兔60只,采用改良Hulth法建立兔膝骨关节炎模型。根据处理方法不同分为空白对照组(n=20)、对照组(n=20)及观察组(n=20)。空白对照组不采取任何方法处理,对照组灌服生理盐水治疗,观察组采用医用臭氧治疗。采用酶联免疫吸附试验测定三组治疗前后TNF-α、IL-1β、MMP-1水平,比较三组治疗效果。结果观察组大白兔采用医用臭氧进行6周处理后关节软骨光泽明亮,光滑无裂纹,滑膜为白色、光泽、光滑,滑液量相对较少,无色;对照组新西兰大白兔局部软化,部分大白兔出血溃疡,滑膜充血、水肿,并且滑液量相对较多。三组处理前TNF-α、IL-1β、MMP-1水平比较差异无统计学意义(P>0.05);观察组处理后TNF-α、IL-1β、MMP-1水平低于对照组和空白对照组(P<0.05),对照组处理后TNF-α、IL-1β、MMP-1水平低于空白对照组(P<0.05)。结论采用医用臭氧治疗兔膝骨关节炎效果理想,能降低关节滑液中TNF-α、IL-1β、MMP-1水平。 Objective To observe the application of medical ozone in rabbit knee osteoarthritis and to investigate its effect on the levels of tumor necrosis factor(TNF-α), interleukin(IL-1β) and matrix metalloproteinase-1(MMP-1) in synovial fluid. Methods 60 New Zealand white rabbits were collected, and the model of rabbit knee osteoarthritis was established by modified Hulth method. They were assigned to the blank control group(n=20), the control group(n=20)and the observation group(n=20) according to the different treatment methods. The blank control group was not given any treatment method, the control group was orally given normal saline, and the observation group was treated with medical ozone. The level of TNF-α, IL-1β and MMP-1 in the three groups before and after treatment were measured by enzyme-linked immunosorbent assay(ELISA). The curative effect was compared among three groups. Results The rabbits in the observation group were treated with medical ozone for 6 weeks. The articular cartilage was bright and smooth without cracks. The synovium was white, shiny, and smooth. The amount of synovial fluid was relatively small and colorless. The local part was softened in the New Zealand rabbits in the control group. Hemorrhage ulcer, synovial congestion and edema could be observed in some rabbits, and the amount of synovial fluid was relatively much; there was no statistically significant difference in the levels of TNF-α, IL-1β and MMP-1 among three groups before the treatment(P〉0.05); the levels of TNF-α, IL-1β and MMP-1 in the observation group after treatment were lower than those in the control group and the blank control group(P〈0.05). The levels of TNF-α, IL-1β and MMP-1 in the control group after the treatment were lower than those in the blank control group(P〈0.05). Conclusion The therapeutic effect of medical ozone in the treatment of knee osteoarthritis is ideal, which can decrease the levels of TNF-α, IL-1βand MMP-1 in synovial fluid.
