白细胞介素-4、干扰素-γ及核因子-κB p65在解淀粉芽孢杆菌致BALB/c小鼠哮喘模型中的表达 被引量:5

Expression of interleukin-4,interferon-γ and nuclear factor-κB p65 in BALB/c mice with asthma induced by Bacillus amyloliquefaciens

摘要 目的探讨白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)与核因子-κB(NF-κB)p65在解淀粉芽孢杆菌致BALB/c小鼠哮喘模型中的表达及意义。方法 15只无特定病原体级BALB/c小鼠随机分为对照组、卵蛋白(OVA)组、解淀粉芽孢杆菌组(细菌组),每组5只。对照组小鼠以生理盐水致敏/激发;OVA组小鼠以0.2 g·L^(-1)OVA混合液致敏,并以25.0 g·L^(-1)OVA激发;细菌组小鼠以1013cfu·L^(-1)解淀粉芽孢杆菌致敏/激发。酶联免疫吸附试验测定各组小鼠血清IL-4、IFN-γ水平;免疫组织化学法检测小鼠肺组织中NF-κB p65表达。结果 OVA组和细菌组小鼠肺组织中NF-κB p65表达显著高于对照组(P<0.01),细菌组和OVA组小鼠肺组织中NF-κB p65表达比较差异无统计学意义(P>0.05)。3组小鼠血清IL-4、IFN-γ水平及IL-4/IFN-γ比较差异有统计学意义(F=90.59、435.09、117.37,P<0.01);OVA组和细菌组小鼠血清IFN-γ水平显著低于对照组(P<0.01),IL-4水平和IL-4/IFN-γ显著高于对照组(P<0.01);细菌组与OVA组小鼠血清IL-4、IFN-γ水平及IL-4/IFN-γ比较差异均无统计学意义(P>0.05)。直线相关分析显示,OVA组小鼠血清IL-4水平与IFN-γ水平呈负相关(r=-0.87,P<0.01);肺组织中NF-κB p65蛋白表达与血清IL-4水平呈正相关(r=0.91,P<0.01),与血清IFN-γ水平呈负相关(r=-0.93,P<0.01)。细菌组小鼠血清IL-4水平与IFN-γ水平呈负相关(r=-0.92,P<0.01);肺组织中NF-κB p65蛋白表达与血清IL-4水平呈正相关(r=0.97,P<0.01),与血清IFN-γ水平呈负相关(r=-0.81,P<0.01)。结论 IL-4、IFN-γ在哮喘发病中起作用,而NF-κB p65表达变化可影响IL-4、IFN-γ水平,NF-κB p65在哮喘炎症调控中发挥重要作用。 Objective To investigate the significance of the expression of interleukin-4( IL-4),interferon-γ( IFN-γ) and nuclear factor-κB( NF-κB) p65 in BALB/c mice with asthma induced by Bacillus amyloliquefaciens. Methods Fifteen BALB/c mice of specific pathogen free were randomly divided into control group,egg protein( OVA) group and Bacillus amyloliquefaciens group( bacteria group),five mice in each group. The mice in control group were sensitized and stimulated with normal saline. The mice in OVA group were sensitized with OVA mixed liquor,and stimulated with 25. 0 g·L^-1VA. The mice in bacteria group were sensitized and stimulated with Bacillus amyloliquefaciens. The levels of serum IL-4 and IFN-γ of mice in the groups were detected by enzyme linked immunosorbent assay. The expression of NF-κB p65 in the lung tissues of mice was detected by immunohistochemistry. Results The expression of NF-κB p65 in lung tissue of mice in OVA group and bacteria group was significantly higher than that in control group( P〈0. 01),but there was no significant difference in the expression of NF-κB p65 in lung tissue of mice between OVA group and bacteria group( P〈0. 05). There was significant difference in the levels of serum IL-4,IFN-γ and IL-4/IFN-γ among the three groups( F = 90. 59,435. 09,117. 37; P〈0. 01). The level of serum IFN-γ in OVA group was significantly lower than that in control group( P〈0. 01),but the levels of serum IFN-γ and IL-4/IFN-γ in OVA group were significantly higher than those in control group( P〉0. 01). There was no significant difference in the levels of serum IL-4,IFN-γand IL-4/IFN-γ between bacteria group and OVA group( P〉0. 05). Linear correlation analysis showed that the serum IL-4 level was negatively correlated to the level of serum IFN-γ( r =- 0. 87,P〈0. 01),the expression of NF-κB p65 protein in lung tissue was positively correlated to the level of serum IL-4( r = 0. 91,P〈0. 01),and the expression of NF-κB p65 protein in lung tissue was negatively correlated to the level of serum IFN-γ in OVA group( r =- 0. 93,P〈0. 01). The level of serum IL-4 was negatively correlated to the level of serum IFN-γ( r =- 0. 92,P〈0. 01),the expression of NF-κB p65 protein in lung tissue was positively correlated to the level of serum IL-4( r = 0. 97,P〈0. 01),and the expression of NF-κB p65 protein in lung tissue was negatively correlated to the level of serum IFN-γ in bacteria group( r =- 0. 81,P〈0. 01). Conclusion IL-4 and IFN-γ play an important role in the pathogenesis of asthma,and the change of NF-κB p65 expression can affect the level of IL-4 and IFN-γ,and NF-κB p65 plays an important role in the regulation of asthma inflammation.
