慢性阻塞性肺疾病患者SIRT1蛋白表达与急性加重风险的相关性研究 被引量:3

Correlation between the SIRT1 Expression and Exacerbation Risk in Patients with Chronic Obstructive Pulmonary Disease

摘要 目的:探讨慢性阻塞性肺疾病(简称慢阻肺)患者外周血单个核细胞(PBMCs)中沉默信息调节因子2相关酶1(SIRT1)蛋白表达与急性加重风险的关系。方法:选择2014年5月至2015年5月在我院诊断为慢阻肺患者30例,分为慢阻肺低风险组和高风险组患者各15例,并与20名健康非吸烟者对照比较。采用Western blot法检测三组研究对象PBMCs中SIRT1蛋白表达。结果:与对照组(0.75±0.06)相比,慢阻肺低风险组患者PBMCs中SIRT1蛋白表达(0.49±0.09)显著降低(P<0.05);与慢阻肺低风险组患者相比,高风险组患者SIRT1蛋白表达(0.28±0.09)进一步降低(P<0.05);Pearson直线相关法分析慢阻肺患者SIRT1蛋白表达水平与FEV1%呈显著正相关(r=0.757,P<0.05),与急性加重频率呈显著负相关(r=-0.776,P<0.05)。结论:慢性阻塞性肺疾病患者SIRT1蛋白表达显著降低,与急性加重频率密切相关。 Objective: To explore the correlation between the silent information regulator factor 2 related enzyme 1 (SIRT1) protein expression in peripheral blood mononuclear cells (PBMCs)and exacerbation risk in patients with chronic obstructive pulmonary disease(COPD). Methods: Thirty COPD patients recruited from our hospital during May 2014 to May 2015 were divided into COPD low risk group and COPD high risk group. Each group contained 15 patients. The control group contained 20 healthy no-smoking subjects. The SIRT1 protein expression in PBMCs was measured with Western blot method. Results: Compared with control group (0.75± 0.06), the SIRT1 protein expression was significantly reduced in PBMCs of low risk COPD patients (0.49± 0.09, P 〈0.05 ). Compared with COPD low risk group, the SIRT1 protein expression was also obviously decreased in PBMCs of high risk COPD patients (0.28± 0.09, P〈0.05 ). Pearson line correlation revealed that the SIRT1 protein was positively correlated with FEVI% predicted value (r=0.757, P〈 0.05 )and negatively correlated with the exacerbation frequency (r=-0.776, P 〈0.05 ). Conclusions: The protein expression of SIRT1 in PBMCs in COPD patients is obviously decreased, which is closely correlatd with exacerbation frequency.
