银屑病患者皮肤组织信号转导和转录激活因子3基因的表达及其意义 被引量:1

Expression and Significance of Signal Transducer and Activator of Transcription 3 in Psoriatic Skin

摘要 目的:检测寻常型银屑病患者皮损区及非皮损区信号转导和转录激活因子3(STAT3)m RNA的表达差异,并探讨其与银屑病皮损发生的关系。方法:采用实时定量PCR方法检测13例寻常型银屑病患者皮损区和非皮损区组织中STAT3基因的m RNA表达水平,并用免疫组织化学方法同时检测两种磷酸化STAT3蛋白在皮损区、非皮损区的表达,比较其差异。结果:寻常型银屑病患者皮损区STAT3基因的m RNA表达水平明显高于非皮损区(P值为0.0001)。两种磷酸化STAT3蛋白均主要表达在角质形成细胞的细胞核,皮损区的表达水平均明显高于非皮损区(P均<0.05),且均可见在真皮浅层血管内皮细胞、淋巴细胞中的表达。结论:STAT3基因在银屑病皮损的角质形成细胞中的m RNA表达显著上调,两种磷酸化STAT3蛋白在皮损区的表达水平均高于非皮损区,提示其参与了银屑病的病理生理过程。 Objective: To investigate the expression of signal transducer and activator of transcription 3 (STAT3) in the skin of psoriatic patients and its relationship with psoriasis. Methods: The mRNA expression level of STAT3 in lesional and non-lesional skin of 13 psoriatic patients were evaluated using real-time quantitative PCR technique while the expressions of two kinds of phosphorylated STAT3 were detected by immunohistochemistry. Results: Compared with non-lesional psoriatic skin, the mRNA expression level of STAT3 was significantly higher in the lesional psoriatic skin (P=0.0001). Immunohistochemistry showed stronger expression of both two kinds of phosphorylated STAT3 in nucleus of keratinocytes in lesional skin relative to non-lesional psoriatic skin and showed high expression in dermal microvascular endothelial cells and surrounding lymphocytes. Conclusion: The mRNA expression of STAT3 and the expression ofphosphorylated STAT3 were significantly higher in the lesional psoriatic skin than non-lesional psoriatic skin, implying that STAT3 might be involved in the pathogenesis of psoriasis.
