摘要 目的研究HSPA1L蛋白在H_2O_2诱导SH-SY5Y细胞损伤中的作用。方法将SH-SY5Y细胞用不同浓度的H_2O_2(1 200、800、600、400、200、100、50、0μmol/L)处理24 h,倒置显微镜观察SH-SY5Y细胞形态学的变化,MTT法检测SH-SY5Y细胞存活率,Western-blot检测SH-SY5Y细胞HSPA1L蛋白表达水平的变化。结果与0μmol/L组比较,不同浓度H_2O_2处理后,细胞形态发生了剂量依赖性损伤;SH-SY5Y细胞的存活率随H_2O_2浓度升高,呈现剂量依赖性降低;HSPA1L蛋白的表达水平随H_2O_2浓度升高,呈现剂量依赖性升高。结论 HSPA1L在H_2O_2诱导的SH-SY5Y细胞损伤模型中,呈现剂量依赖性升高,这可能是细胞应对氧化应激损伤的一种补偿机制。 Objective To investigate the effect of HSPA1 L expression on SH-SY5 Y cell damage induced by hydrogen peroxide. Methods SH-SY5 Y cells were treated with hydrogen peroxide at different concentrations( 1 200,800,600,400,200,100,50 and 0 μmol / L) for 24 h. The morphological changes of SH-SY5 Y cells were observed by inverted microscope. The cell viability was measured by MTT method. The expression of HSPA1 L was detected by western blot. Results Compared with the control group, hydrogen peroxide impaired the cell morphology in a dose-dependent manner. Meanwhile,the cell viability of SH-SY5 Y cells was significantly decreased and HSPA1 L expression was increased in SH-SY5 Y cells when treated with hydrogen peroxide. Conclusion The expression of HSPA1 L protein is increased in SH-SY5 Y cells when treated by hydrogen peroxide, which may be one of the compensating mechanisms in response to the oxidative damage.
