摘要
目的研究D5S818、D8S1179、D7S820、CSF1PO、D2S1338、D3S1358、vWA、D21S11、D16S539、PentaE、TPOX、TH01、D19S433、D18S51、FGA、D6S1043、D13S317、D12S391等18个短串联重复序列(short tandem repeats,STR)基因座在甘肃地区东乡族人群的遗传多态性。方法采用PCR扩增及毛细管电泳技术对615名甘肃地区东乡族个体18个STR基因座进行分析。结果 615名甘肃地区东乡族个体在18个STR基因座上,共检出213种等位基因,850种基因型,其分布均符合Hardy-Weinberg平衡(P〉0.05),18个基因座的杂合度(H)在0.633~0.928之间,匹配概率(Pm)在0.013~0.205之间,个体识别概率(DP)在0.795~0.987之间,多态信息含量(PIC)在0.560~0.920之间,非父排除概率(PE)值在0.332~0.854之间。结论实验研究了甘肃地区东乡族人群18个STR基因座的遗传多态性,为人类群体遗传学及法医学后续研究提供详实可靠的基础数据。
Objective This study was to determine the frequencies and parameters of Dongxiang population 18 short tandem repeat loci(D5S818、D8S1179、D7S820、CSF1PO、D2S1338、D3S1358、vWA、D21S11、D16S539、PentaE、TPOX、TH01、D19S433、D18S51、FGA、D6S1043、D13S317、D12S391)in Gansu region.Methods PCR amplification and capillary electrophoresis technologies were employed to determine the genetypes of 18 STR loci for 615 unrelated Dongxiang individuals.Results 213 alleles and 850 genotypes were recognized in Dongxiang population from Gansu region.The results demonstrated that all loci were found to be no deviation from Hardy-Weinberg equilibrium(P〉0.05).Heterozygosity(H) ranged from 0.633 to 0.928,matching probability(Pm) ranged from 0.013 to 0.205,power of discrimination(DP) ranged from 0.795 to 0.987,polymorphism information content(PIC) ranged from 0.560 to 0.920,power of exclusion(PE) ranged from 0.332 to 0.854.Conclusion This work studied the genetic polymorphism of 18 short tanderm repeat loci of Dongxiang population in Gansu region.The resulting genetic data to provide basic data for subsequent human population genetics and forensic DNA research work.
