集中空调系统水中嗜肺军团菌脉冲场凝胶电泳指纹图谱分析 被引量:1

Analysis of fingerprinting of Legionella pneumophila strains in water from centralized air conditioning system by pulsed field gel electrophoresis

摘要 目的 研究某市2014年公共场所集中空调系统水中嗜肺军团菌的分子指纹图谱,初步建立某市嗜肺军团菌的脉冲场凝胶电泳(PFGE)分型数据库。方法 对某市2014年公共场所集中空调系统水分离的34株嗜肺军团菌,采用PFGE技术进行电泳,并经Bio Numerics 6.6软件进行聚类分析。结果34株嗜肺军团菌的PFGE图谱共分为24种PFGE带型,不同地区嗜肺军团菌存在同样的酶切图谱,同一采样场所不同冷却塔检出菌株的PFGE带型相同,聚类分析结果显示菌株间相似性系数为23.80%-100.00%。结论 某市2014年公共场所集中空调系统水嗜肺军团菌菌株基因型散在分布,呈多态性和复杂性,同时具有克隆化特征。 Objective To identify and analyze the molecular types of Legionella pneumophila strains isolated from water in Centralized Air Conditioning System of public places of a city in 2014, and develop the PulseNet Database of Legionella pneumophila. Methods Thirty-four strains of Legionella pneumophila were analyzed by Pulsed field gel electrophoresis (PFGE). The cluster results of PFGE were analyzed by BioNumerics 6.6. Results Thirty-four strains of Legionella pneumophila were divided into twenty-four PFGE patterns. Some of Legionella pneumophila strains from different districts had same PFGE pattern, while different cooling towers from one district had same PFGE pattern. Cluster diagram showed the LP1 strains similarity coefficients were 23.80%-100.00%. Conclusion Legionellapneumophila strains isolated from cooling water of public places not only had polymorphism and complexity in distribution, but also had cloncal characteristic.
