缺氧诱导因子-1α和11-19赖氨酸富集白血病基因相关因子2在前列腺癌组织的表达及其临床意义 被引量:2

Expression and clinical significance of hypoxia inducible factor -1 α and eleven -nineteen lysinerich leukemia-associated factor 2 in prostate cancer

摘要 目的检测前列腺癌及前列腺增生组织中癌基因缺氧诱导因子-1α(HIF-1α)和抑癌基因11-19赖氨酸富集白血病基因相关因子2(EAF2)的表达,探讨两者与前列腺癌的临床病理特征的相关性。方法应用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)法检测48例前列腺癌及30例前列腺增生组织HIF-1α和EAF2基因的表达,并对前列腺癌患者血清前列腺特异抗原(PSA)、Gleason评分、TNM分期及骨转移与生存时间等临床病理特征进行研究。结果HIF-1α和EAF2在前列腺癌组织中阳性率分别为72.9%和39.6%,在前列腺增生组织中阳性率分别为43.3%和63.3%,差异有统计学意义(P〈0.05)。EAF2表达与PSA水平:PSA≤20ng/ml:68.8%(11/16),PSA〉20ng/ml:25.0%(8/32);Gleason评分:〈7分:50.0%(8/16),=7分:64.7%(11/17),〉7分:6.7%(1/15);TNM分期:Ⅰ-Ⅱ期:66.7%(12/18),Ⅲ-Ⅳ期:23.3%(7/30);骨转移:无:51.7%(15/29),有:15.8%(3/19)呈负相关(P〈0.05)。HIF-1α表达则相反。两者表达均与年龄无明显相关。本组前列腺癌患者生存期的比较显示EAF2表达呈阳性的患者生存期明显高于EAF2呈阴性的患者(P〈0.05)。结论前列腺癌组织中癌基因HIF-1α表达增强,抑癌基因EAF2表达下调,其表达强度与肿瘤的生物学特征密切相关。 Objective To detect the expression of oncogenes hypoxia inducible factor -1α (HIF- 1α) and tumor suppressor genes eleven - nineteen lysinerich leukemia - associated factor 2 (EAF2) in prostate cancer tissues and to investigate the correlation between them and the clinicopathologi- cal features of prostate cancer. Methods The expression of HIF -1α and EAF2 in 48 cases of prostate cancer and 30 cases of benign prostatic hyperplasia tissue was detected by streptavid - in - peroxidase (SP) immunohistochemical method, and serum prostate specific antigen (PSA), Gleason score, TNM stage, bone metastasis, survival time and other clinicopathological parameters in prostate cancer patients were clinically analyzed. Results The positive rate of HIF - let and EAF2 in prostate cancer tissues was 72. 9% and 39. 6%, and that in benign prostatic hyperplasia was 43. 3% and 63.3%, respectively, with the difference being statistically significant (P 〈 0. 05). The EAF2 expression in prostate cancer was nega- tively correlated with PSA [PSA ≤20 ng/ml, 68.8% (11/16) ; PSA 〉20 ng/ml, 25.0% (8/32) ], Gleason score [ 〈7, 50.0% (8/16); =7, 64.7% (11/17); 〉7, 6.7% (1/15)], TNM staging [ Ⅰ~Ⅱ, 66.7% (12/18);Ⅲ~Ⅳ, 23.3% (7/30)], and bone metastasis [non bone metastasis, 51.7% (15/29) ; bone metastasis, 15.8% (3/19) ] (P 〈0. 05). The HIF- 1α expression had the op- posite trend. Both of them were not related to age. The survival time in protate cancer patients positive for the EAF2 expression was significantly longer than in those negative for the EAF2 expression ( P 〈 0.05 ). Conclusion HIF - 1 α expression was enhanced and EAF2 expression was down - regulated in prostate cancer tissues, which is closely related to the biological characteristics of the tumor.
