基于单壁金属有机纳米管(SWMONTs)的药物传递系统(英文)

Drug Delivery System Based on Single-wall Metal-organic Nanotubes(SWMONTs)

摘要 功能化单壁金属有机纳米管(SWMONTs)可以装载抗癌药物5-氟尿嘧啶(5-FU).材料表征结果表明,药物与SWMONTs的存在比例为每1.0 g载体SWMONTs中含有0.33 g的5-FU.释放曲线表明,在PBS缓冲液(p H=7.4)中24 h释放量为79.6%,而在缓冲液(p H=5.7)中24 h释放量为75%.细胞4T1和RAW264.7被用于细胞毒性实验.2种癌细胞的最高致死率均发生在最高实验浓度的载药体系(5FU@SWMONTs)处理时间最长的样品中.结果表明,5FU@SWMONTs对癌细胞的毒性会因为SWMONTs组分的存在而增强. A self-assembled functionalized metal-organic nanotubes, SWMONTs could be loaded with the anticancer drug 5-Fluorouracil (5-FU). The characterization of the material indicated the presence of drug associated with SWMONTs (5FU@SWMONTs) in a proportion of 0.33 g 5-FU per 1.0 g of SWMONTs. The drug demonstrated a slow release profile where 79.6% of the drug was released in PBS buffer (pH7.4) in 24 h, while 75% in PBS buffer (pH5.7) in 24 h. The cytotoxic tests were carried out against two cell lines: 4T1 and RAW264.7. The highest lethality of cancer cells occurred at the highest concentration of 5FU@SWMONTs for the longest period of time. The results demonstrate that the cyto- toxicity of 5FU@SWMONTs to cancer cells was improved by the SWMONTs component.
