摘要
目的 探讨上皮细胞黏附分子(EpCAM)在非小细胞肺癌(NSCLC)中的表达及临床意义.方法 采用实时定量聚合酶链反应(Real-time PCR)法和免疫组织化学EnVision染色法分别检测64例NSCLC手术患者配对癌组织和远端正常组织中EpCAM mRNA和蛋白的表达水平,并分析其与临床病理特征的相关性.结果 NSCLC癌组织中EpCAM mRNA的表达水平显著高于远端正常组织(P<0.01),64例配对癌组织中,44例EpCAM mRNA表达水平上调,上调表达的EpCAMmRNA表达与NSCLC的肿瘤大小和肿瘤分期显著相关(P<0.05).免疫组织化学结果显示,EpCAM蛋白在NSCLC癌组织中阳性表达率较高,为59.38% (38/64),与远端正常组织中阳性表达率(15.63%,10/64)比较有显著的相关性.且EpCAM蛋白表达与NSCLC的肿瘤大小和肿瘤分期显著相关(P<0.05).结论 EpCAM在NSCLC发展过程中起着重要的作用,并有望成为抗NSCLC治疗的一个潜在生物学标志.
Objective To investigate the expression pattern of epithelial cell adhesion molecule (EpCAM) in non-small cell lung carcinoma (NSCLC) and its clinical significance.Methods EpCAM mRNA and protein expression was detected in 64 paired tumor tissues and normal adjacent tissues from NSCLC patients by quantitative real-time quantitative polymerase chain reaction (Real-time PCR) and immunohistochemistry,respectively.Results EpCAM mRNA expression in tumor tissues was significantly higher than that in paired normal adjacent tissues (P < 0.01).Up-regulated EpCAM mRNA expression was detected in 44 NSCLC patients and the higher level of EpCAM mRNA expression was dramatically correlated with tumor size and TNM stage (P < 0.05).The immunohistochemistry results showed that EpCAM protein positive rate in tumor tissues was also higher (59.38%,38/64) than that in normal adjacent tissue,and the elevated EpCAM protein expression was significandy correlated with NSCLC tumor size and TNM stage (P < 0.05).Conclusion EpCAM plays an important role in the development of NSCLC,and it is expected to be a potential biological marker for anti NSCLC therapy.
