2型糖尿病大鼠气阴两虚夹瘀证动物模型的建立对大鼠肾脏功能的影响 被引量:4

摘要 目的:研究2型糖尿病大鼠气阴两虚夹瘀证动物模型对大鼠肾脏功能的影响。方法:将40只SPF级雄性SD大鼠随机分为空白组10只和造模组30只,空白组基础饲料喂养,造模组高脂饲料喂养,造模组共筛选得到24只符合气阴两虚夹瘀证的2型糖尿病大鼠,将造模大鼠分为模型组和糖脉康组,每组各12只。所有实验大鼠均参照人体换算给药剂量5倍浓度灌胃给药,糖脉康组给予糖脉康颗粒给药,空白组和模型组给予同体积换算的生理盐水,连续给药3周。检测其血清肌酐、尿素氮、尿蛋白、肾脏湿质量等指标,判断该动物模型的建立对大鼠肾脏功能影响。结果:药物干预后,糖脉康组与模型组比较,血清肌酐、尿素氮有不同程度下降趋势;尿蛋白含量明显低于模型组,有显著性差异(P<0.01)。模型组大鼠肾湿质量、脏器指数大于空白组,并具有显著性差异(P<0.01),糖脉康干预后,肾脏湿质量、脏器指数相对于模型组有下降,具有显著性差异(P<0.01)。结论:气阴两虚夹瘀证模型大鼠的肾功能存在损伤,糖脉康对改善肾功能具有一定的作用。 Objective:To study the influence of establishment of animal model of type Ⅱ diabetic rats with qi-yin deficiency with blood stasis syndrome on renal function in rats. Methods:Forty SPF-grade male SD rats were randomly divided into the blank group and the model group,with 10 and 30 cases respectively in each group. The ones in the blank group were fed with basic diet, while the ones in the model group were fed with high fat diet. Twenty-four rats in the model group which belonged to type Ⅱ dia- betic rats with qi-yin deficiency with blood stasis syndrome were screened, and divided into the model group and Tangmaikang (TMK) group, with 12 rats in each group. All the experimental rats were treated with the drug which had 5 times dosage's density by gavage with reference to body conversion. The ones in TMK group were treated with Tangmaikang Granule (TMKG) ,while the ones in the blank group and the model group were given the same volume of normal saline for 3 weeks, serum creatinine, urea nitrogen ,urinary protein, pathology, kidney dampness quality and other indicators were detected to determine the impact of the establishment of the animal model on renal function in rats. Results:After drug intervention, compared with the model group, TMK group's serum creatinine and urea nitrogen decreased to some degree,and its urinary protein contents were markedly lower than that in the model group, and there were significant differences ( P 〈 0.01 ). Conclusion : The renal function of type Ⅱ diabetic rats with qi-yin deficiency with blood stasis syndrome has damage,and TMK has certain effect on the improvement of renal function.
