摘要
设计一对能区分经典株与变异株的PRRSV特异性引物,建立PRRSV经典株与变异株的鉴别诊断方法,用该方法对2013年实验室收集到的来自131个免疫PRRS弱毒疫苗猪场的493份临床送检样品进行RT-PCR检测。PRRSV的检出率为27.2%(134/493),其中变异株的检出率为23.1%(114/493),经典株的检出率为8.11%(40/493),20份样品可以同时检出PRRSV变异株和经典株,检出率为4.1%(20/493)。对临床送检的30个免疫PRRS活疫苗猪场的62个猪群1 079份血清,用IDEXX PRRS X3抗体检测试剂盒进行了血清学分析,结果显示总体阳性率为91.01%(982/1079),育肥猪群阳性率最高,达95.26%;其次为母猪群,阳性率高达93.7%;保育猪相对较低,为82.72%。根据猪群临床症状和PRRS状况将猪群分为四种类型。对62个免疫PRRS弱毒疫苗的规模化猪场进行了抗体分析,PRRSV抗体的离散度超过40%的猪场达59.7%(37/62)。这些结果说明,变异株为主要流行毒株,同时存在经典株与变异株的混合感染;免疫猪群抗体不整齐,目前弱毒疫苗免疫效果不理想。
PRRSV specific primers and identify primers (the distinction between the classical strains and HP-PRRSV) were designed respectively. In 2013, our laboratory detected 493 clinical samples by using RT-PCR method for PRRSV epidemiological investigation. The results showed that the PRRSV positive rate was 27.2% (134/493), while HP-PRRSV positive rate was 23.1% (114/493), the classic strains positive rate was 8.11% (40/493), part of the pig farms can be simultaneously detected with the HP-PRRSV and classical strains (20/493)of one sample. It shows the co-existence of HP- PRRSV with classical strains, however, HP-PRRSV was superiority pandemic strain. Furthermore, we tested 1079 serum samples with IDEXX PRRS X3 Antibody Test Kit for PRRSV serological analysis, it shows that the overall PRRSV antibody positive rate was 91.01% (982/1 079). Positive rate of growing and fattening pigs is the highest, 95.26%. Followed by the sow group, 93.7%, and the nursery pigs are the lowest, 82.72%. According to the clinical symptoms, PRRS pigs can be divided into four types. The analysis of 62 large-scale pig farms shows the coefficient of variation of sow herds PRRSV antibody of 59.7% (37/62) pig farms is over 40%. The survey results show PRRS is still quite active and unstable in most of the herds.
