D-手性肌醇及D-松醇对缓解HepG2细胞胰岛素抵抗的作用 被引量:9

摘要 对D-手性肌醇及D-松醇缓解胰岛素抵抗作用进行评价,选用HepG2人肝癌细胞建立胰岛素抵抗细胞模型。将实验分为正常对照组和D-手性肌醇及D-松醇实验组,分别设立50、100、200、400mg/L4个剂量。采用四甲基偶氮唑盐法(MTT法)检测受试样品对细胞增殖的影响。选用1×10^-6mol/L浓度的胰岛素诱导细胞建立胰岛素抵抗模型,给予不同浓度D-手性肌醇及D-松醇,考察二者对胰岛素抵抗HepG2细胞葡萄糖消耗量的影响。实验结果表明,在200-1000 mg/L的浓度范围内D-手性肌醇和D-松醇对HepG2细胞的正常增殖无显著影响。D-手性肌醇浓度为50、100和200 mg/L时,对胰岛素抵抗HepG2细胞的葡萄糖消耗量与模型对照组相比有显著促进作用(p〈0.05),浓度为400 mg/L时,有极显著性差异(p〈0.01);D-松醇浓度为200 mg/L时,对胰岛素抵抗HepG2细胞的葡萄糖消耗量与模型对照组相比有显著促进作用(p〈0.05),浓度为400 mg/L时,有极显著性差异(p〈0.01)。因此,D-手性肌醇和D-松醇均能够缓解HepG2细胞胰岛素抵抗现象,且在1000 mg/L浓度范围内对HepG2细胞正常增殖无显著影响。 The effects of D-chiro-inositol and D-pinitolon mitigation of insulin resistance in Hep G2 cells was tested. The cells were divided into normal control group and D-chiro-inositol / D-pinitol dosage groups(50,100,200,400 mg/L). MTT test was chosen to inspect the toxicity of the D-pinitol and D-chiro-inositol. Hep G2 cells were given 1×10^-6mol/L insulin in order to establish an insulin resistance model. The results suggested that D-chiroinositol in 50,100 and 200mg/L promoted the glucose intake signifi cantly(p〈0.05),and D-chiro-inositol in 400mg/L promoted the glucose intake very signifi cantly(p〈0.01). D-pinitol in 200mg/L increased the glucose intake signifi cantly(p〈0.05); There was very signifi cant difference between insulin resistance control group and 400mg/L D-pinitol treated group(p〈0.01). In conclusion,D-chiro-inositol and D-pinitol have the ability to mitigate insulin resistance in Hep G2 cell and have no signifi cant effect on proliferation of Hep G2 cell within the concentration of 1000mg/L.
