摘要
目的:观察三叶青乙酸乙酯提取物对结肠癌HT29细胞皮下移植瘤的诱导凋亡作用,探讨其可能的作用机制,为三叶青在临床上应用于结肠癌的治疗提供实验依据。方法:以HT-29结肠癌细胞荷瘤裸鼠为模型。实验分5组,模型组、阳性对照组及三叶青提取物低、中、高剂量组,各组裸鼠给予相应药物或生理盐水灌胃14 d。观察各组裸鼠的一般情况,以及对HT29细胞皮下移植瘤的抑瘤率;HE染色后观察肿瘤组织细胞形态学改变,免疫组织化学方法检测瘤组织内Caspase-3的蛋白表达水平。结果:三叶青提取物中、高剂量组及阳性组瘤重较模型组减轻(P<0.05),阳性组瘤重最轻;中药各组随着剂量的增加,抑瘤率上升。与模型对照组比较,各给药组荷瘤鼠瘤细胞凋亡程度增强;三叶青提取物中、高剂量组瘤组织Caspase-3蛋白表达水平显著上调。结论:三叶青乙酸乙酯提取物可抑制结肠癌HT29细胞皮下移植瘤的生长,作用机制之一为上调Caspase-3蛋白的表达,诱导结肠癌HT29细胞皮下移植瘤凋亡。
Objective: To study the apoptosis- inducing effect of Ethylacetate extracts of Tetrastigma hemsleyanum on colorectal cancer cell HT29 subcutaneous transplanted tumor and then to explore the possible mechanism and provide the experimental basis for tumor therapy with Tetrastigma hemsleyanum. Methods: Nude mice bearing colorectal cancer with HT29 cells were selected as models. Experiment was divided into 5 groups,model,positive( 5- Fu) and low,middle,high dose Tetrastigma hemsleyanum extract groups,nude mice in each group were given the corresponding medicine or saline for 14 days,then the general situation of each groups,was observed and the tumor inhibition rate on HT29 cells subcutaneous transplantation tumor was calculated; morphologic changes of tumor tissue cells in each groups were observed by HE stained,Caspase- 3 protein expression in tumor tissue was determined by immunohistochemical method. Results: The tumor weight of middle,high dose Tetrastigma hemsleyanum groups and 5- Fu group was lighter than that in model group mice( P〈0. 05),the tumor weight of positive group was the lightest; the tumor inhibition rate of the three Tetrastigma hemsleyanum extract groups was raised with increasing of dosage. The apoptosis degree in each medicine group was obvious to compare with model group by observing the morphologic changes of tumor cells. The Caspase- 3 expression of HT- 29 tumor- bearing nude mice in middle and high dose Tetrastigma hemsleyanum extract groups was significantly up- regulated to compare with model group. Conclusion: Tetrastigma hemsleyanum ethylacetate extract can inhibit the growth of colon cancer HT29 cells subcutaneously transplanted tumor; up- regulating Caspase-3 protein expression and inducing HT29 cells appoptosis was one of the mechanism.
