摘要
目的:探讨长链非编码RNA(long noncoding RNA,lncRNA)中的反义缺氧诱导因子(antisense hypoxia inducible factor,a HIF)和位于细胞周期激酶抑制因子4(INK4)基因座中反义非编码RNA(antisense noncoding RNA in the INK4 locus,ANRIL)在胃癌组织与血浆中的表达。方法:收集20例胃癌患者癌组织及对应的癌旁组织、33例胃癌患者和20例健康者血浆,采用实时荧光定量PCR测定胃癌组织和血浆中lncRNA a HIF和ANRIL的表达水平,分析lncRNA a HIF、ANRIL在血浆中的表达与胃癌组织表达的相关性。结果:胃癌组织中lncRNA a HIF和ANRIL的表达水平均明显高于癌旁组织(t分别为-4.463,-4.886,P均<0.01);胃癌患者血浆中lncRNA a HIF和ANRIL的表达水平均明显高于健康者(t分别为-4.232,-4.450,P<0.01),且与胃癌组织中的表达水平呈正相关(r分别为0.536,0.524,P<0.05)。结论:lncRNA a HIF和ANRIL在胃癌组织和患者血浆中均高表达,且血浆与组织中的表达水平呈正相关。
Objective : To explore the expression level of antisense hypoxia inducible factor (aHIF) of long noncoding RNA (lncRNA) and antisense noncoding RNA (ANRIL) which located in cell cycle kinase inhibitory factor 4 (INK4) loci in gastric cancer tissue and plasma. Methods: We collected 20 cases gas- tric cancer tissues and the normal tissue adjacent to carcinoma of gastric cancer patients, 33 cases plasma of patients with gastric carcinoma and 20 cases plasma of healthy subjects. Real-time fluorescence quantitative PCR was used to determine the mRNA expression levels of lncRNA aHIF and ANRIL; the relationship be- tween the two mRNA expressions in plasma and tissues of gastric cancer patients were analyzed. Results: The mRNA expression levels of lncRNA aHIF and ANRIL in gastric cancer tissue were obviously higher than those of adjacent carcinoma tissues (P 〈0.01 ). Compared with the healthy subjects, the mRNA ex- pression of aHIF and ANRIL in plasma of gastric cancer patiens was significantly increased ( P 〈 0. 01 ), which positively related to the mRNA expression of gastric cancer tissues ( r= 0. 536,0. 524, P 〈 0.05 ). Conclusion : LncRNA aHIF and ANRIL were overexpression in tissue and plasma of gastric cancer patients.
