α-细辛醚调控GADD153和Smac mRNA表达诱导人食管癌细胞株Eca-109凋亡研究 被引量:5

Experimental study onα-Asarone-induced apoptosis in human esophageal carcinoma Eca-109 cell line I by regulating GADD153 and Smac mRNA expression levels

摘要 目的α-细辛醚具有止咳、平喘、解痉、镇静、抗惊厥等功效,文中旨在探讨α-细辛醚对人食管癌细胞株Eca-109促凋亡作用,以及对生长抑制DNA损伤基因153(GADD153)和第2个线粒体来源的半胱氨酸酶的激活剂(Smac)表达的影响。方法体外培养人食管癌Eca-109细胞,以加入药物不同分为空白对照组、5-氟尿嘧啶组(5-FU,500μg/m L)和不同剂量的α-细辛醚(25、50、100μg/m L);继续培养后,采用MTT法检测细胞增殖率,Annexin V-PI法检测细胞凋亡率,Western blot和实时荧光定量PCR法分别检测GADD153、Smac蛋白及mRNA的相对表达量。结果 5-氟尿嘧啶组、α-细辛醚25μg/m L组、α-细辛醚50μg/m L组、α-细辛醚/100μg/m L组细胞凋亡率[(15.59±0.48)%、(4.61±1.13)%、(9.48±1.09)%、(12.41±0.98)%]较空白对照组[(0.29±0.08)%]明显升高(P<0.05),与空白对照组比较,α-细辛醚组细胞GADD153、Smac蛋白及GADD153、Smac mRNA相对表达量均升高,差异有统计学意义(P<0.05)。结论α-细辛醚通过上调GADD153及Smac mRNA的表达,抑制人食管癌Eca-109细胞增殖,促进其凋亡,凋亡途径由内质网和线粒体通路共同介导。 Objective αAsarone has the effect of relieving cough and asthma as well as sedative, hypnotic and anticonvulsive function. Our study was designed to explore the effects of apoptosis induced by α Asarone on human esophageal carcinoma Eca109 cell line as well as the expression levels of GADD153 and Smac mRNA. Methods Human esophageal carcinoma Eca109 cells were cultured in vitro, which were divided into blank control group, 5FU group( 500μg/mL) andαAsarone group of different dosages ( 25,50,100μg/mL) . After cultivation, MTT method and Annexin VPI were used to measure cell proliferation rate and apoptosis rate. Expression levels of GADD153 and Smac protein and mRNA were detected by western blotting and real time quantitative PCR. Results Compared with blank control group, the cell proliferation rates in other groups decreased significantly (P〈0.01), and cell apoptosis rate increased significantly ( P〈0.01) . Compared with blank control group, the expression levels of GADD153 and Smac protein and mRNA in other groups increased significantly( P〈0.05) . Conclusion α Asarone can inhibit cell proliferation and induce the apoptosis of human esophageal carcinoma Eca109 by regulating the expression levels of GADD153 and Smac gene.
