Determination of anthraquinone in tea by GPC-HPLC

摘要 建立了用凝胶渗透色谱-高效液相色谱测定茶叶中蒽醌的方法。以乙酸乙酯、乙酸乙酯-正己烷(1∶1,V/V)液液萃取,凝胶渗透色谱净化,收集时间为11~17 min,以甲醇+0.5%H3PO4溶液(65+35,V/V)为流动相,ZORBAX SB-C18分离,外标法定量。蒽醌质量浓度在10~200μg/L范围内线性关系良好,相关系数r为0.9993,检出限为6μg/kg,定量限为10μg/kg。茶叶样品在10,50,180μg/kg 3个加标浓度下,蒽醌的回收率为73.2%~97.5%,相对标准偏差为4.0~7.9%。 A method was developed for determination of anthraquinone in tea based on high performance liquid chromatography(HPLC) with gel permeation chromatography(GPC). Sample was extracted with ethyl acetate,ethyl acetate- n-hexane(1: 1,V/V),cleaned up with GPCwas and collected at 11 ~ 17 min. The analyte was then separated by HPLC ZORBAX SB-C18 column using methanol /0. 5% phosphoric acid(65: 35,V/V) as mobile phase,and quantified by the external standard method. The calibration curve was linear in the range of10 ~ 200 μg/L with correlation coefficient of 0. 9993. The limit of detection(LOD) was 6 μg/kg and the limit of quantitation(LOQ) was 10 μg/kg. Average recoveries of the method at three different spiked levels ranged from73. 2 % to 97. 5% with the relative standard deviation(RSD) from 4. 0% to 7. 9%.
