红花水煎液对博莱霉素诱导的硬皮病小鼠肺组织胶原含量的影响 被引量:4

摘要 目的:观察红花水煎液对博莱霉素诱导的硬皮病小鼠肺组织的HE和MASSON染色病理变化及MASSON染色胶原含量分布变化,探讨红花对硬皮病肺纤维化的影响。方法:将32只BALB/C小鼠随机分为4组:空白对照组、模型对照组、醋酸泼尼松组、红花组,每组8只。除空白对照组外,各组建立硬皮病小鼠模型。醋酸泼尼松组予醋酸泼尼松混悬液0.2 m L灌胃,红花组予红花水煎液0.2 m L灌胃,空白对照组和模型对照组予同体积生理盐水灌胃。采用HE和MASSON染色观察各组小鼠肺的病理改变,使用IPP 6.0图像分析软件测定MASSON染色胶原含量分布变化,SPSS 17.0软件进行统计分析。结果:与空白对照组比较,模型对照组小鼠肺组织胶原含量显著升高(P<0.05);与模型对照组比较,醋酸泼尼松组和红花组小鼠肺组织胶原含量明显降低(P<0.05);醋酸泼尼松组与红花组胶原含量比较,差异无统计学意义(P>0.05)。结论:红花能够有效降低博莱霉素诱导的硬皮病小鼠肺组织胶原含量,具有抗硬皮病肺纤维化的作用。 Objective:To probe the effects of safflower decoction on pulmonary fibrosis by observing the staining pathological changes of HE and MASSON and the distribution of MASSON staining collagen content in bleomycin-induced mice with scleroderma.Methods:Thirty-two BALB/C mice were randomly divided into 4 groups:a blank control group,a model control group,a prednisone group and a safflower group,8 rats in each. Except for the blank control group,mouse models with scleroderma were established in other groups.Rice in the Prednisone group were given intragastric administration with 0.2 mL prednisone suspension,while those in the safflower group were given safflower decoction in the same way.Mice in the blank control group and the model control group were also given intragastric administration with saline of he same volume.HE and MASSON stainings were used to observe the pathological changes of lung in each group.IPP6.0 image analysis software was used to determine the distribution of MASSON staining collagen content,and SPSS17.0 software was used for statistical analysis.Results:Compared with the blank control group,the collagen content in lung tissue of mice in the model control group significantly increased ( P 〈 0.05 ) .Compared with the model control group,the collagen content in lung tissue of mice in the prednisone group and the safflower group significantly decreased ( P 〈 0.05 ). The difference of collagen content between the prednisone group and the safflower group was not statistically significant ( P 〉 0.05 ) Conclusion:Safflower can effectively reducethe lung collagen content in bleomycin-induced mice with scleroderma,with a function of against pulmonary fibrosis.
