摘要
目的 建立同时测定鸦胆子药材中1,2-油酸-3-亚油酸甘油酯和甘油三油酸酯两种脂溶性成分含量的方法。方法 采用UPLC-ELSD法,色谱柱为Thermo Accucore XL C18柱(250 mm×2.1 mm,4μm);流动相为乙腈-二氯甲烷(体积比70∶30);流速为0.4 m L·min-1;柱温为30℃;进样量为4μL;漂移管温度为45℃;雾化器载气压为0.276 MPa。结果 1,2-油酸-3-亚油酸甘油酯和甘油三油酸酯进样量分别在0.509~2.55μg(r=0.999 6)和0.685~3.42μg(r=0.999 1)内线性关系良好,平均加样回收率分别为97.5%和99.5%,RSD分别为1.7%和2.3%(n=9)。结论 建立的方法可用于同时测定鸦胆子中1,2-油酸-3-亚油酸甘油酯和甘油三油酸酯的含量,为控制鸦胆子药材质量提供试验依据。
Abstract : Objective To develop a method for the simultaneous determination of the contents of 1,2-olein-3- linolein and glycery trioleate in Brucea javanica by UPLC-ELSD. Methods The separation was carried out on a Thermo Accueore XL C18 colurrm(250 mm ×2. 1 mm,4 μm) with a mobile phase of acetonitrile and dichloromethane ( V: V = 70: 30)at a rate of 0. 4 mL·min- 1 ;the column temperature was 30 ℃ and the ELSD tube temperature was set at 45 ℃. Results The liner range of calibration curve was 0. 509 - 2. 55 μg ( r = 0.999 6 ) and 0.685 - 3.42 μg ( r = 0.999 1 ) for 1,2-olein-3-1inolein and glycery trioleate respectively. The average recovery was 97.5% and 99.5% with RSDs of 1.7% and 2.3% ( n = 9 ), respectively. Conclusions The method can be applied for the simultaneous determination of 1,2-olein-3-1inolein and glyc- ery trioleate in Brucea javanica, and provide a reference for the quality control of Brucea javanica.
