摘要
以具有代表性的三种二肽Gly-Gln,Ala-Gln和Gly-Gly为氮源,以葡萄糖为糖源,制备了三种二肽Amadori中间体(APs).采用HPLC监控反应进程以比较三种二肽的反应活性,其顺序依次为Ala-Gln>Gly-Gln>Gly-Gly.采用LC-MS/MS和ESI-MS/MS对三种二肽的APs的纯度及断裂模式进行表征和分析,结果表明,三种二肽APs的LC/MS纯度分别可达95%(Gly-Gln-APs)、97%(AlaGln-APs)和98%(Gly-Gly-APs),三种二肽APs断裂模式相似,主要碎片离子包括丢失糖分子形成的[M-162]+和[M-180]+碎片离子,以及丢失水分子形成的[M-36]+和[M-84]+离子.加香应用评价表明,三种二肽APs皆能与烟香谐调,提高香气量和烟气浓度,其中Ala-Gln-APs添加效果最好,对卷烟的香气特征和口感特性皆有明显的正面作用.
Three dipeptide Amadori intermediates( APs) were prepared by three representative dipeptide GlyGln,Ala-Gln and Gly-Gly as the nitrogen source,and glucose as sugar sources. The reaction process was monitored by HPLC to compare reactivity of dipeptides,the reactivity order is Ala-Gln Gly-Gln Gly-Gly.The purity and fracture modes of three dipeptide APs were characterized and analysis using LC-MS / MS and ESI-MS / MS. The results showed that the LC / MS purity of three dipeptide APs was up to 95%( Gly-GlnAPs),97%( Ala-Gln-APs) and 98%( Gly-Gly-APs). Three fracture pattern of dipeptide APs was similar,the main fragment ions including loss of sugar molecules formed [M-162]+and [M-180]+fragment ions; and loss of water molecule is formed [M-36]+and [M-84]+ions. The cigarette application evaluation showed that the three dipeptide APs have encountered with incense smoke harmonize and improve the aroma and the amount of gas concentration,wherein Ala-Gln-APs shows the best effects. An obvious positive effect could be found both in aroma and taste characteristics.
