摘要
目的研究木犀草素联合替莫唑胺对脑胶质瘤大鼠VEGF表达的影响。方法建立大鼠U87-MG脑胶质瘤模型,以随机数字表法分为四组,每组20只。接种14d后给予替莫唑胺组替莫唑胺25mg/(kg·d),木犀草素组木犀草素20mg/(kg·d),联合组给予木犀草素20mg/(kg·d)、替莫唑胺25mg/(kg·d),均配置为2m L生理盐水溶液后腹部注射,空白组每天给予等量生理盐水。给药5周后将脑胶质瘤细胞切片染色,免疫组化法检测各组大鼠脑胶质瘤细胞中VEGF水平。结果联合组VEGF阳性与强阳性表达数量之和所占比例为20%,与空白组、木犀草素组、替莫唑胺组85%(χ2=16.942)、70%(χ2=10.101)、55%(χ2=5.227)相比均明显下降(P<0.05)。结论木犀草素联合替莫唑胺治疗U87-MG脑胶质瘤,可有效抑制肿瘤VEGF表达,对脑胶质瘤的化疗效果起增敏作用。
Objective To explore the effect of luteolin combined with temozolomide for the expression of VEGF in rat brain glioma. Methods Rat U87-MG glioma model was established. By the random number table method, the rats were divided into four groups with 20 rats in each. After inoculation with 14d, temozolomide 25mg/(kg · d) was used in the temozolomide group, luteolin 20mg/(kg · d) was used in the luteolin group, luteolin 20mg/(kg · d) and temozolomide 25mg/(kg · d) were used in combined group, all with abdominal injection of 2mL saline solution, and equivalent normal saline was used in the blank group every day. After administration with 5 weeks, the glioma ceils were stained sections, immunohistochemical method was used to detect the level of VEGF in rat glioma cells in each group. Results The proportion of sum of VEGF positive and strong positive expression quantity of combined group was 20%. Compared with the blank group, luteolin group, temozolomide group 85%(x2=16.942),70%(x2=10.101), 55%(x2=5.227), it was significantly decreased (P 〈 0.05). Conclusion Luteolin combined with temozolomide in treatment of U87-MG brain glioma could effectively inhibit the expression of VEGF in tumor, and it played the role of sensitization on the effect of chemotherapy on brain glioma.
