摘要
目的 观察大鼠急性脑梗死后β-淀粉样蛋白(Aβ)的变化规律以及延迟肢体缺血后处理对其的干预作用,探讨延迟肢体缺血后处理对大鼠急性脑梗死的保护作用及机制。方法 采用轮流夹闭双下肢股动脉主干,2min缺血/2min再灌注,各5个周期,连续5d进行延迟肢体缺血后处理。比较各组大鼠右侧脑梗死体积以及运用免疫组织化学染色检测各组Aβ的表达。结果 在第1d,单纯再灌注组与延迟肢体缺血后处理组上述各项指标几乎相等。但从第2~5d,单纯再灌注组脑梗死体积均明显高于后者,差异有统计学意义(P〈0.05);与延迟肢体缺血后处理组各时间点比较,单纯再灌注组Aβ的阳性细胞数均明显较高,差异有统计学意义(P〈0.05)。延迟肢体缺血后处理组与单纯再灌注组Aβ均在第1d就开始增多,第2d达高峰,但高峰值后者明显高于前者。结论 延迟肢体缺血后处理可能抑制大鼠脑梗死体积扩大,其机制可能与抑制脑梗死后Aβ的表达有关。
Objectives We analyzed the change law of Aβ on acute cerebral ischemia and explored the effect of delayed limb ischemia on cerebral ischemia in rats. Methods The double femoral artery was cliped by turns 2min ischemia /2min reperfusion, 5 cycles and 5 consecutive days. The right cerebral infarct volume and the expression of Aβ were de tected by immunohistochemical staining. Results At the first day, the infarct volumes and the expression of Aβ were almost equal in the simple reperfusion group and delayed limb ischemia group. From the second day to the fifth day, the infarct volume of the simple reperfusion group was significantly higher than that of delayed limb ischemia group (P 〈0.05). Compared with the simple ischemia reperfusion group, the number of Aβ positive cells in delayed limb ischemia group was low at each time point (P〈0.05). Aβ positive ceils increased at the first day both in t delayed limb ischemia group and the simple reperfusion group, which reached the peak at the second day, and began to fall at the third day. The peak value in delayed limb ischemia group was significantly higher in the simple reperfusion group. Conclusions Delayed limb ischemia may inhibit the expansion of infarct size, and its mechanism may be related to inhibition of Aβ expression.
