摘要
目的:应用联合应激法建立单纯心理应激小鼠实验模型。方法:雄性昆明小鼠30只,随机分为2组,分别为心理应激(psychological stress,PS)组和正常对照(normal control,NC)组。适应环境1周后,连续给予PS组小鼠隔离应激30 d,并每天随机给予捕食应激或社会失败应激1次;NC组群居饲养。末次应激后第2天进行旷场实验,断颈取血,通过酶联免疫吸附试验(enzyme-linked immuno-sorbent assay,ELISA)检测血清中促肾上腺皮质激素释放激素(corticotropin releasing hormone,CRH)、促肾上腺皮质激素(adrenocorticotropic hormone,ACTH)、皮质醇(cortisol,CORT)水平,并与NC组进行比较。结果:PS组经联合心理应激后,5 min旷场活动明显减少(P<0.01)。PS组与NC组比较,血清中CRH、ACTH及CORT水平均明显升高,差异具有统计学意义(P<0.01)。结论:通过联合应激法,可建立一种可靠的单纯心理应激动物实验模型。
Objective：To build the mice model of pure psychological stress based on combined stimuli. Methods：Thirty male mice were randomized into 2 groups：psychological stress group（n =15） and control group（n =15）. After one-week acclimation,the mice model of psychological stress was subjected to social isolation stress（30 days）,and randomized to predator stress（1/day） or social defeat stress（1/day）. Open-field test was performed at 24 h after the last psychological stress. Then,the average contents of corticotropin releasing hormone（CRH）,adrenocorticotropic hormone（ACTH）and cortisol（CORT） detected by ELISA and were compared between two groups. Results：The activity was significantly reduced in open-field test in psychological stress group（P〈0.01）. The average contents of CRH,ACTH and CORT were significantly higher in psychological stress group than in control group（P〈0.01）.Conclusion：It is a credible method to establish animal model of pure psychological stress through combined stimuli.
