摘要
目的:探讨三七多糖对2型糖尿病大鼠肝脏高同型半胱氨酸代谢信号通路3个关键分子亚甲基四氢叶酸还原酶(MTHFR)、胱硫醚β-合酶(CBS)、5-甲基四氢叶酸高半胱氨酸甲基转移酶(MTR)表达的影响。方法:将60只雄性SD大鼠随机分为空白组、模型组以及三七多糖治疗组,每组20只,模型组以及三七多糖治疗组给予高脂高糖饮食6个月建立2型糖尿病模型,模型建立成功后三七多糖治疗组给予三七多糖57 mg/(kg·d)灌胃治疗,模型组给予同体积生理盐水灌胃,分别在治疗4周和8周后处死大鼠各半,收集血清和肝脏进行相关指标的检测。结果:治疗4周后大鼠血糖以及胰岛素抵抗指数三七多糖治疗组较模型组降低(P<0.05),8周后进一步降低,胰岛素水平治疗4周后三七多糖治疗组较模型组降低(P<0.05),8周后较4周无进一步改善;三七多糖治疗后,大鼠叶酸较模型组有一定升高,但差异无统计学意义(P>0.05),维生素B12在治疗4周后较模型组明显升高(P<0.05),治疗8周后进一步升高;同型半胱氨酸在治疗4周后明显降低(P<0.05),但在8周后无进一步改善;治疗4周后,与模型对照组比较,三七多糖治疗组MTHFR、MTR以及CBSm RNA及蛋白表达升高(P<0.05),治疗8周后进一步升高,但未达到空白组水平。结论:三七多糖可显著降低模型大鼠血清中同型半胱氨酸含量,其可能是通过调控高同型半胱氨酸信号通路相关蛋白MTHFR、CBS、MTR的表达来实现。
Objective: To explore the influence of panax polysaccharide on the expressions of three key molecules MTHFR, CBS, MTR of homocysteine signaling pathway of the rats with type two diabetes mellitus (T2DM). Methods: Sixty rats were randomized into the blank group, the model group and the treatment group of panax polysaccharide, 20 rats each group, the model group and the treatment group of panax polysaccharide were given high fat and high sugar diet for six months to establish T2DM rat models, after successfully establishing the models, the treatment group of panax polysaccharide accepted intragastric administration of panax polysaccharide in the concentrations of 57mg/(kg·d), the model group received physiological saline in the same volume, the rats were sacrificed after treating in four weeks and eight weeks respectively, the related indexes were detected after collecting the serum and liver. Results: Blood glucose and insulin resistance index of the treatment group of panax polysaccharide decreased obviously compared with the model group after treating in four weeks (P〈0.05), these indexes lowered after treating in eight weeks, insulin level decreased after treating in four weeks when the treatment group ofpanax polysaccharide was compared with the model group notably (P〈0.05), there was no improvements after treating in eight weeks compared with these in four weeks; after treating with panax polysaccharide, the levels of folic acid of the rats rose compared with the model group, but there was no statistical meaning (P〉0.05), the levels of vitamin B12 improved compared with the model group after treating in four weeks (P〈0.05), they improved further after treating in eight weeks; the level ofhemocysteine decreased obviously after treating in four weeks (P〈0.05), while there was no improvements after treating in eight weeks; compared with the model group after treating in four weeks, mRNA and protein expressions of MTHFR, MTR and CBS rose evidently in the treatment group of panax polysaccharide (P〈 0.05), these expressions improved after treating in eight weeks, but they didn't reach the level of the blank group. Conclusion: Panax polysaccharide could notably decrease the contents of homocysteine in the rat model apparently, which might be realized by regulating the expressions of MTHFR, MTR and CBS in homocys- teine signaling pathway
