氢气预处理对离体缺氧-复氧胸主动脉的保护作用

Protective effect of hydrogen pretreatment on hypoxia-reoxygenation isolated thoracic aorta

摘要 目的:观察氢气对缺氧-复氧离体小鼠胸主动脉组织结构和功能的保护作用。方法:90只ICR小鼠随机分为6组(n=15):分别为内皮完整未通氢对照组(Con+E)、去内皮未通氢对照组(Con-E)、内皮完整预通氢组(Pre H_2+E)、去内皮预通氢组(Pre H_2-E)、内皮完整再通氢组(Post H_2+E)、去内皮再通氢组(Post H_2-E)。血管环缺氧10min后复氧以制备缺氧-复氧模型,氢气干预在再通氢组于复氧后7 min开始进行,在预通氢组于缺氧前10 min开始。经生物信号采集与分析系统测定血管环张力的变化,观察血管组织及细胞结构的改变。结果:在缺氧-复氧后,去内皮预通氢组的血管张力保持稳定,而去内皮未通氢对照组的血管张力明显降低(P<0.01)。组织细胞形态结构结果显示预通氢处理的两组血管组织细胞形态均良好,仅局部肌丝紊乱;再通氢处理的两组血管出现轻度损伤:肌细胞局灶性溶解、局部肌丝紊乱或消失、核周隙增宽;而未通氢对照的两组血管则均出现明显的内膜剥离等改变,肌细胞弥漫性溶解和线粒体空泡变、内质网扩张明显、核膜皱缩明显、核周隙增宽、细胞凋亡明显。结论:氢气预处理具有维持缺氧-复氧后血管张力稳定及保护组织细胞形态结构的作用,其效果明显优于损伤后的氢气再干预。
