摘要
目的探讨金钱草总黄酮提取液抑制大鼠草酸钙结石形成机制的研究。方法常规饲养大鼠1周,采用乙二醇饮水和氯化铵灌胃造模草酸钙结石大鼠模型,将30只SD大鼠随机分为空白对照组(A组)、草酸钙结石模型组(B组)和金钱草总黄酮组(C组)。给药4周后收集各组大鼠24 h尿液,检测尿p H值及24 h尿钙、尿草酸、尿酸、尿葡胺聚糖和尿凝血酶原片段-1的含量。结果金钱草总黄酮组大鼠24 h尿量较草酸钙结石模型组增加,差异有统计学意义(P<0.05),与空白对照组比较差异无统计学意义(P>0.05)。金钱草总黄酮组大鼠与草酸钙结石模型组大鼠比较,其24 h尿钙和尿草酸含量均降低,而尿凝血酶原片段-1的含量增加,差异均有统计学意义(P<0.05)。而三组间尿p H值、尿酸经比较均无明显差异(P>0.05)。结论金钱草总黄酮提取液可能通过增加大鼠24 h尿量,降低尿液中成石因子-尿钙和尿草酸的含量,增加尿液中抑石因子-尿凝血酶原片段-1含量,从而抑制草酸钙结石的形成。
Objective To discuss the mechanism of the total flavone extract of lysimachia on calcium oxalate stone for- mation in rats. Methods Conventional breeding rats for a week, the rat model of calcium oxalate stone was made by us- ing ethylene glycol drinking water and ammonium chloride. 30 adult male SD rats were randomized to three groups, the control group (group A), the stone group (group B), and the total flavone extract of lysimachia group (group C). All rats were bred 4 weeks, the 24 hours' urine of rats were collected, and then the pH value, the calcium, oxalate acid, uric acid, the GAGs and the prothrombin fragment-1 of the urine were detected. Results The 24 hours" urine volume of the total flavone extract of lysimachia group were increased than the stone group, there was significant difference (P〈0.05), while there was no significant difference compared with the control group (P〉0.05), and the calcium, oxalate acid of the urine of the total flavone extract of lysimachia group were reduced, and the urinary prothrombin fragment-1 increased compared with the stone group(P〈0.05), while there was no significant difference in pH value and uric acid of the urine among three groups (P〉0.05). Conclusion The total flavone extract of lysimachia can increase the 24 hours' urine vol ume, and can decrease the promoting stone factors-calcium, oxalate acid of the urine, can increase the urinary pro- thrombin fragment-l, thereby inhibiting the formation of calcium oxalate stones.
