急性心肌梗死致应激性高血糖大鼠模型的建立 被引量:3

Establishment of rat model with stress hyperglycemia induced by acute myocardial infarction

摘要 目的建立急性心肌梗死(AMI)致应激性高血糖大鼠的实验动物模型。方法 40只SD雄性大鼠随机均分为四组。A、B、C组结扎左冠状动脉前降支,分别于缺血15、30和45min后再灌注120min;S组为假手术对照。持续监测各组心电图和缺血前(T0)、缺血末(T1)和再灌注末(T2)时的血糖,检测T2时血促肾上腺皮质激素(ACTH)、皮质酮和胰岛素水平,2,3,5-氯化三苯基四氮唑(TTC)染色分析心肌梗死面积,观察模型成功及大鼠死亡的比例。结果 A、B、C组大鼠结扎前降支后均出现了显著的ST-T改变,再灌注后可见病理性Q波,TTC染色出现白色梗死区。与S组比较,A、B、C组T1、T2时的血糖和T2时的ACTH、皮质酮、胰岛素水平均升高(P<0.05),且B、C组高于A组(P<0.05)。B、C组模型成功比例高于A组(P<0.05),B组和C组分别有1只和3只大鼠死亡。结论成功建立AMI致应激性高血糖实验动物模型;缺血30min再灌注120min的模型稳定,病死率低。 Objective To establish an experimental rat model with stress hyperglycemia induced by acute myocardial infarction(AMI).Methods Forty male SD rats were equally and randomly divided into 4 groups.The rats in groups of A,B and C experienced myocardial ischemia for 15,30 and 45 minutes,respectively,by the ligation of left anterior descending coronary artery,which was followed by reperfusion for 120 minutes.Sham operation was performed in group S.ECG was monitored continuously.Blood glucose(BG)was detected before ischemia(T0),at the end of ischemia(T1)and reperfusion(T2).Serum levels of adrenocorticotropic hormone(ACTH),corticosterone and insulin were detected at T2.Myocardial infarction area was measured by 2,3,5-triphenyl tetrazolium chloride(TTC)staining.The numbers of rats with successful models and death were recorded.Results Obvious changes of ST-T segment and Q wave were seen on ECG and white infarcted area occurred showed by TTC staining in groups of A,B and C.Compared with group S,BG at T1 and T2 and ACTH,CS and insulin at T2 were increased in groups of A,B and C(P〈0.05),which were higher in groups of B and C than those in group A(P〈0.05).The proportion of successful rat model was more in groups of B and C than that in group A(P〈0.05).One rat in group B and three rats in group C were died.Conclusion The rat model with AMI-induced stress hyperglycemia has been established successfully.Myocardial ischemia for 30 minutes followed by reperfusion for 120 minutes is suitable to establish a stable rat model of AMI-induced stress hyperglycemia with lower mortality.
