摘要
目的探索小鼠骨髓间充质干细胞(BMSCs)的培养和其肝硬化模型的制备,以及两者之间的相互作用。方法首先,提取小鼠骨髓,移至培养瓶中,根据全骨髓贴壁法的原理培养干细胞;然后,采用CCl4为主的方法对小鼠进行肝硬化模型的制备;最后,把培养成功的骨髓干细胞通过肝动脉和尾静脉途径移植治疗造模成功的肝硬化小鼠,观察疗效。结果根据多种单克隆抗体的表达,使用流式细胞仪进行鉴定,成功培养小鼠骨髓间充质干细胞;对造模小鼠肝脏组织进行病理切片,病理报告图示典型的肝硬化,证明造模成功;通过肝动脉和尾静脉途径移植治疗肝硬化模型小鼠,最后报告干细胞治疗的病理结果,肝硬化程度减轻,说明骨髓间充质干细胞具有一定的疗效。结论自体骨髓干细胞移植治疗肝硬化是有效的,但仍需要基础研究和大样本的临床数据。
Objective To explore the cultivation of mice bone marrow mesenchymal stem cells and the preparation of mice liver cirrhosis model, as well as the interaction of two methods. Methods First of all, the mice BMSCs were extracted to culture bottle, further separated and cultured in vitro; then, mice liver cirrhosis model was prepared by the approach of CCl4-based; finally, BMSCs were transplanted into mice liver cirrhosis model through the hepatic artery and tail vein. Results According to the expression of monoclonal antibodies, the mice BMSCs were identified by means of flow cytometry and cultured successfully; mice liver tissue was executed via pathological section, the pathological findings reported typical cirrhosis and modelled successfully; BMSCs transplantation therapy had a certain effect was proved by final pathology results and cirrhosis abated. Conclusion It is effective that autologous BMSCs transplantation treat liver cirrhosis, but basic research and a large sample of clinical data are still demanded.
