表没食子儿茶素没食子酸酯对大鼠肾草酸钙结石模型成石的影响

摘要 目的观察不同剂量表没食子儿茶素没食子酸酯(EGCG)对大鼠肾结石模型成石的影响。方法 50只雄性SD大鼠分为5组,A组为空白对照组;B组为单纯成石组,采用1%乙二醇及2%氯化铵进行配制成的造石液2ml灌胃;C、D及E组均为EGCG干预组,分别在B组处理方法的基础上每天予10、30及50 mg/kg的0.1%EGCG溶液灌胃处理;均连续进行28 d。取右肾组织测定丙二醛(MDA)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平;左肾冰冻切片,偏光显微镜观察结晶情况。结果 B组较A组肾组织MDA浓度显著增高,而SOD、GSH-Px、CAT浓度显著降低(均P<0.01)。与B组比较,C、D及E组肾组织MDA水平降低及SOD水平增加(均P<0.01)。肾组织结晶与EGCG治疗剂量呈负相关(r=-0.483,P<0.01)。结论过氧化损伤是导致高草酸诱导的肾小管上皮细胞损伤的主要机制和结石形成的主要途径;EGCG可减轻机体的过氧化损伤,降低形成肾结石的危险性。
