摘要
目的:研究复脉汤对大鼠缺血性心律失常Cx43 Ser279/282磷酸化位点的作用机制。方法:将SD大鼠分为假手术组、模型组、美托洛尔组、炙甘草汤组,复脉汤高、中、低剂量组,灌胃给药14 d后,观察各组大鼠Ⅱ导联心电图,结扎SD大鼠冠状动脉左前降支(除假手术组外),制成缺血性心律失常模型,记录缺血性心律失常出现时间以及发生次数,术后采用蛋白免疫印迹法(Western blot)测定各实验组大鼠心肌细胞Cx43 Ser279/282位点磷酸化水平。结果:复脉汤各剂量组及美托洛尔组能显著延迟心律失常出现时间,减少心律失常发生次数。复脉汤各剂量组、美托洛尔组均可不同程度地降低Cx43 Ser279/282磷酸化水平。结论:复脉汤对缺血性心律失常的保护作用可能基于降低Cx43 Ser279/282磷酸化水平。
Objective:To study the mechanism of Fumai decoction on ischemic arrhythmias. Methods:70 SD rats were divided into sham operation group,model group,metoprolol tartrate group,Zhigancao decoction group,Fumai decoction high,medium and low dose group. Rat models of ischemic arrhythmia were induced by left anterior descending artery ligation except sham group. Then occurrence time,numbers of ischemic arrhythmia were recorded. The expression of Cx43 Ser 279/282 in myocardial tissue was detected by Western blot. Results:Compared with model group,arrhythmia occurrence time was delayed,numbers of ischemic arrhythmia,phosphorylated expression of Cx43 Ser279/282 were decreased in Fumai decoction groups and metoprolol group. Conclusion:Fumai decoction has the protective effect on ischemic arrhythmias.Its mechanism may be related to reducing phosphorylated expression of Cx43 Ser279/282.
